有氧运动与营养干预对2型糖尿病大鼠糖、脂代谢影响的研究

【摘要】 目的:建立较为符合2型糖尿病特点的大鼠模型,观察有氧运动(aerobicexercise)和补充葛根素对2型糖尿病大鼠糖、脂肪代谢紊乱和胰岛素抵抗的影响,探讨有氧运动及营养干预治疗2型糖尿病的可行性。 方法:SD大鼠70只,空白对照组采用普通饲料喂养,DM建模组采用高脂高糖饲料持续喂养4周辅以腹腔注射小剂量(45mg/kg)链脲佐菌素(STZ)的方法,建立比较符合人类2型糖尿病特征的SD大鼠模型,成模的大鼠随机分为4组,即DM对照组、DM运动干预组、DM营养干预组、DM运动+营养干预组。采取有氧运动或/和葛根素干预的方法进行干预。干预前后观测体重,空腹血糖(FBG)、血清胰岛素(INS)、甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白(HDL-C)、和低密度脂蛋白(LDL-C)含量的变化,计算胰岛素敏感指数(ISI)。 结果:高脂高糖饲料喂养辅以腹腔注射小剂量STZ的方法,可以建立比较符合人类2型糖尿病特征的SD大鼠模型。实验结果表明:通过有氧运动或/和葛根素的干预,各组糖尿病大鼠空腹血糖值(FBG)、总胆固醇(TC)、甘油三酯(TG)、自由脂肪酸(FFA)、低密度脂蛋白(LDL-C)值较之实验前相比呈显著性下降(P<0.05),血清胰岛素(INS)和高密度脂蛋白(HDL-C)值有显著性升高(P<0.05)。有氧运动和营养干预后,与DM对照组相比,DM运动+营养干预组大鼠的TC、TG、FFA、FBG值出现大幅的下降,有非常显著性差异(P<0.01),LDL-C值出现小幅的下降(P<0.05),而HDL-C、ISI、INS值出现大幅的升高,有非常显著性差异(P<0.01)。与空白对照组相比,除了DM运动+营养干预组的INS有显著性的差异(P<0.05),FBG、HDL-c、LDL-c、TG、TC和FFA值都没有显著性的差异(P>0.05),说明DM运动+营养干预组的糖、脂代谢水平已经接近空白对照组,而且其ISI有非常显著性的提高(P<0.01),说明糖尿病大鼠的胰岛素敏感性得到提高,胰岛素抵抗得到改善。因此,可以看出,通过有氧运动和营养干预,能很好的改善2型糖尿病大鼠的糖、脂代谢,消除胰岛素抵抗现象,从而达到彻底治疗糖尿病的目的。

【Abstract】 Obejective: To establish the SD rats model conforms to 2 diabetes characteristics, To observe 2 diabetes rats sugar fat metabolism disorder and the insulin resistance influence by aerobic exercise and the Puerarin supplemented. And to discuss the scientific of 2 diabeteses theories by aerobic exercise and the medicine supplemented.Methods: SD rats were modeled into T2DM rats by high quality of heat feeding for 4weeks and abdominal injection of streptozotocin, then were randomly divided into 5groups:In addition to the normal contrast group, The DM model group was given no treatment; the DM sports group, the DM medicine group and the sports medicine group were respectively treated by aerobic exercise, Puerarin and aerobic exercise+Puerarin. The groups were recorded at which were before the beginning of the treatment, 6 treatment weeks later. The level of FBG, serum insulin, serum FFA, serum TG, serumTC, serum HDL-C, serum LDL-C was measured; and insulin sensitivity index(ISI) was calculated. Results: The method was proved practical that SD rats were modeled into T2DM rats by high quality of heat feeding and abdominal injection of streptozotocin. After experiment, The DM rats FBG, TC, TG, LDL-C, FFA have a decline and there is remarkable difference(P<0.05), The INS, HDL-C have a ascend trend, and there is remarkable difference(P<0.05).As far as the group DM sports+DM medicine is concerned, contrasted by DM model group, Its TC、 TG 、 FFA、 FBG and INS have a huge decline and there is very remarkable difference (P<0.01), Its LDL-C have a little decline (P<0.05), While Its HDL-C、 ISI and INS have a huge ascend and there is remarkable difference (P<0.01).contrasted by normal group, Its FBG have a little ascend, Its INS have a little decline (P<0.05), but they are closed to normal value;While its ISI have a huge ascend (P<0.01).Therefore, we can make a conclusion that aerobic exercise and the Puerarin supplemented can ameliorate 2 diabetes rats sugar, the fat metabolism disorder and the insulin resistance influence, and can improve carbohydrate metabolism and lipid metabolism to some extent.
