革叶蕨的离体培养和快速繁殖
Techniques of Rapid Propagation in Vitro of Rumohra Adiantiformis

【摘要】 本研究以三叉蕨科革叶蕨属革叶蕨(Rumohra adiantiformis)为材料,对革叶蕨的离体培养和快速繁殖进行了研究。一、将革叶蕨外植体用不同杀菌剂、不同温度热水浴预处理,探讨减少初代培养污染率的有效措施。二、以初代培养获得的无菌外植体接种在含有一定浓度激素的培养基中,研究不同类型外植体分化能力的强弱并进行脱分化程度的比较。三、最佳分化外植体在正交设计2因素(蔗糖浓度、基本培养基)3水平条件下进行不定芽诱导的试验及最佳pH值的筛选,并进一步研究激素的种类和浓度对不定芽诱导增殖的影响。四、研究不同激素处理对不定芽生根的影响,得到健壮生根组培苗后,再进行不同基质配方对组培苗移栽成活率和生长势影响的试验,得到理想的效果,为获得优质的革叶蕨苗,提供了生产依据。 结果表明: (1)降低初代培养过程中污染率的方法 用3g/L根菌清杀菌剂对外植体进行24h预处理可使无菌率达到96.67%。效果明显好于最佳水浴处理(40℃处理1h)。 (2)不同部位外植体分化能力和脱分化程度的比较 革叶蕨茎段、叶片和叶柄接种在附加一定浓度IAA和NAA的MS培养基上后,均可不同程度的分化出芽,器官分化能力的强弱顺序为:茎尖>茎段>叶柄>叶片;茎尖接种后20d左右产生绿色球状物,经60天培养产生丛生苗,叶柄和叶片出芽慢,在培养30d后切口处先形成少量瘤状结构,40d后切口处形成簇生芽,其中叶片在与叶柄相连的切口处更易于长芽,而较细小的上部或边缘叶片接种后大多褐化,诱导能力弱。 (3)不定芽的诱导增殖方法 试验结果得出,以无菌茎段在蔗糖浓度30g/L、pH值5.80的1/2MS培养基中诱
【Abstract】 In this paper, we focused on the techniques for rapid propagation of Rumohra adiantiformis. Firstly, we searched for the effective measures to reduce the pollution rate in the first generation of the Rumohra, which had already been dealt with different germicide and at different water temperature. Secondly, we studied the ability of the differentiation between different kinds of explants and compared the degrees of the dedifferentiation. We putted the fungus-less explant into the culture medium that had been mixed with some hormone. Thirdly, we carried further study on the types of the hormone and the effective of the hormone concentration on the propagation of adventitious buds. We could get the result by the inducing of the adventitious bud and screening the best PH in the condition of orthogonal design 2 factors in 3 levels. Fourthly, we reasearched on the effects of different hormones on rooting of adventitious buds. After getting healthy plants, we investigated the effection of different groups of substances on the survival rates and growth vigor during domestication. All the work done aim to attain high quality Rumohra and an available method for commercial propagation of large scale in Rumohra adiantiformis . The main results are as followings :(1) The method of reducing the pollution rate during the elementary propagation. Using 3g/L germicide treats explants for 24h has a virus-free rate of 96.67%, its better than that of thermotherapy which treats the explant for 1h at 40℃.(2) Comparisons of differentiation ability in different positions of explants and dedifferentiation degrees with them. Differents organs cultured on the same medium have a differents differentiation ability.
