从免疫角度探讨盐酸川芎嗪对早孕小鼠妊娠影响的机理
Effect of Ligustrazine Hydrochloride on the Mice Fertility and Its Mechanism during Early Pregnancy in View of Immunity

【摘要】 目的:通过检测妊娠中的激素,脾淋巴细胞增殖情况、转化生长因子β1(Transforminggrowth factor β1,TGF-β1)及其受体(Transforming growth factor β1 receptor Ⅰ,TGF-β1RI)表达的情况,探讨妇科临床常用药盐酸川芎嗪做为妊娠禁忌药的机理。 方法:将妊娠小鼠分为高、中、低三组,从妊娠第1天持续灌胃不同浓度川芎嗪(10mg/天,2mg/天,0.4mg/天),持续6天,同时设立对照组,灌胃生理盐水。于妊娠第7天,眼眶取血,同时摘取脾脏、子宫等组织。通过放射免疫法测定血清中的孕酮(Progesterone,P)含量,通过细胞培养及MTT法观察脾细胞增殖情况。通过免疫组化法分析子宫蜕膜中TGF-β1及其受体的表达,通过原位杂交法检测TGF-β1 mRNA表达情况。 结果:高浓度组血清中孕酮浓度与对照组相比,明显增加且差异显著(P<0.01)。对于脾淋巴细胞增殖,低浓度组MTT比色后OD值与对照组基本相同,而中、高浓度组均显著高于对照组(P<0.01),且随川芎嗪剂量增加而增加。TGF-β1、TGF-β1R、TGF-β1mRNA均为胞浆着色,细胞核内无着色。TGF-β1免疫组化着色面积比例高于TGF-β1RI。TGF-β1着色面积也高于TGF-β1mRNA的原位杂交着色面积,三者在子宫内表达部位大致相同。对于TGF-β1,高浓度组与对照组相比,表达量显著降低,具有明显差异(P<0.05)。同时,通过对TGF-β1mRNA检测发现高浓度组与对照组相比,mRNA量也是显著降低,差异显著(P<0.05)。通过对TGF-β1RI检测发现无论是低、中,还是高浓度组,与对照组相比均无明显差异(P>0.05)。 结论:对妊娠小鼠灌胃川芎嗪,可以促进脾淋巴细胞的增殖,可能是因为川芎嗪抑制了TGF-β1的表达,同时孕酮虽然作为一种免疫抑制剂,但是其作用的发挥还需要依赖于包括TGF-β1等的各种细胞因子。
【Abstract】 Object: In order to investigate the mechanism of Ligustrazine Hydrochloride as a taboo for pregnant women.Methods: The pregnant mice were treated intragastrically with Ligustrazine Hydrochloride (10mg/d, 2mg/d or0.4mg/d) for six days from the first day of pregnancy and the control group was treated with saline only. At the seventh day of pregnancy, uterus and spleen were removed and blood was collected. The progesterone level in serum was measured by RIA. The splenocyte were cultured in vitro, and the amount of splenocyte was detected by MTT. The expression of the proteins of TGF-β1 and TGF-β1RI were detected by Immunohistochemical staining and the expression of TGF-β1 mRNA was detected by in situ hybridization histochemistry.Result: The level of progesterone in serum distinctly increased with different concentration of Ligustrazine Hydrochloride as compared to the control group. As to the cultured spleen cells after treated with 10mg/d and 2mg/d of Ligustrazine Hydrochloride, the cell number sharply increased. Ligustrazine Hydrochloride can weaken the expression of TGF-β1, but had no effect on the expression of TGF-β1RI in none of the three experimental groups. The expression pattern of TGF-β1mRNA was similar to the TGF-β1. TGF-β1mRNA was distinctly reduced at the dose of 10mg/d of Ligustrazine Hydrochloride.Conclusion: Ligustrazine Hydrochloride can enhance the multiplication of spleen cells may be by reducing the expression of TGF-β1. Progesterone as a immune inhibitor exert its function-depending on many cytokine include TGF-β1.
