三种中草药制剂调控雄性小鼠体内镉毒性的试验研究

【摘要】 大量研究表明,镉对雄性哺乳动物生殖系统具有明显的毒性,小剂量镉即可导致睾丸精子数减少,附睾内少精或无精,进而影响雄性生殖功能,因此迫切需要找到防治镉中毒的有效的药物。为了探讨中草药绞股蓝总甙、黄芪多糖和甘草甜素能否控制镉对雄性生殖系统的毒性,进行了镉诱导睾丸生殖细胞凋亡的体内控毒作用的实验研究。 研究方法:本实验采用25日昆明龄雄性小鼠110只,体重18~22g,随机分成11组,分别为对照组、氯化镉组、各中草药低、中、高浓度组。通过腹腔注射中草药绞股蓝总甙、黄芪多糖和甘草甜素处理后,再同时腹腔注射氯化镉的在体实验模型。实验结束时取睾丸称重,比较各组体重、睾丸质量及睾丸脏器系数的变化;采用病理组织学方法检测睾丸生殖细胞的形态学变化;采用TUNEL法检测睾丸生殖细胞凋亡率。 实验结果:各中草药拮抗组小鼠睾丸生殖细胞凋亡率明显降低,与氯化镉组相比,差异极显著(P<0.01)。其TUNEL法检测结果为:对照组的睾丸生殖细胞凋亡率为10.51±2.3281,镉组的睾丸生殖细胞凋亡率为36.53±2.2983,绞股蓝总甙各组的睾丸生殖细胞凋亡率为:18.01±1.7465;15.02±4.3901;12.47±1.8139,黄芪多糖各组的睾丸生殖细胞凋亡率为:19.05±1.9589;15.32±4.7612;13.60±3.8207,甘草甜素各组的睾丸生殖细胞凋亡率为:27.48±2.6827;22.15±2.8593:15.51±4.9810。表明绞股蓝总甙、黄芪多糖和甘草甜素对镉引起的睾丸生殖细胞凋亡有一定程度的拮抗作用。 实验结论:1) 绞股蓝总甙、黄芪多糖和甘草甜素具有一定的抑制镉致睾丸生殖细胞凋亡的作用;2) 这三种中草药对镉致睾丸生殖细胞凋亡的作用的大小为:绞股蓝总甙>黄芪多糖>甘草甜素。

【Abstract】 There is the research enunciation, the Cadmium (Cd) have an obvious toxicity to procreative system in the male mammal. A little cadmium could cause testicle germ cell count decrease , epididymal sperm count cut down and vanish, and affect arrenotoky function. For inquirying in to Gypensldes、 Astragalus Polysacharide and Glycyrrizin can control the toxicity of the Cadmium to procreative system , this experiments researches carried on the Cadmium to induce inner the bodies of testicle germ cell apoptosis. Research Methods: This experiment that total 110, the weight 18-22g and 25th age of male mice in Kunming were divided into random 11 groups which the control group, the Cd group and each density groups of low, medium and high of three kinds of the herb medicine. Male mice as object injected with several dose cadmium chloride after celiac injected Gypensldes 、Astragalus Polysacharide and Glycyrrizin. Testicle viscera coefficient was counted, testis was detected by pathological and the apoptosis of testis genital cell were assessed by employing TdT- mediated dUTP-biotin nick end labeling (TUNEL) technique.Research Results: To compare with the Cd group, the difference in every Chinese traditional medicine group was significant (P<0. 01). The Results showed that the apoptosis percent of genital cells in low, midst, high-dose groups of Gypensldes, Astragalus Polysacharide and Glycyrrizin are 18.01 + 1.7465; 15.02±4. 3901; 12. 47±1. 8139; 19.05±1.9589; 15. 32±4. 7612; 13. 60±3.8207; 27. 48 ±2. 5827; 22.15 ±2. 8593; 15. 51 ±4. 9810, whereas the control group and the Cd group are 10. 51±2. 3281, 36. 53±2. 2983. So the result indicate that Gypensldes, Astragalus Polysacharide and Glycyrrizin have protect effects on genital cell apoptosis in mice induced by Cadmium Chloride.Research Conclusion: 1)Gypensldes, Astragalus Polysacharide and Glycyrrizin have protect effects on genital cell apoptosis in mice induced by Cadmium Chloride. 2)The size of function is Gypensldes > Astragalus Polysacharide > Glycyrrizin.
