针刺对更年期大鼠卵巢组织NO、NOS、SOD、MDA及超微结构的影响

【摘要】 目的:探讨针刺“足三里”、“关元”穴对更年期雌性大鼠卵巢组织抗氧化作用及超微结构影响 方法:本实验对更年期雌性大鼠针刺“足三里”、“关元”穴,采用考马斯亮兰测定法测定卵巢组织中蛋白含量;应用硝酸还原酶法测定一氧化氮(NO)含量;应用分光光度法测定一氧化氮合酶(NOS)活力;应用黄嘌呤氧化酶法测定超氧化物歧化酶(SOD)活力;应用硫代巴比妥酸染色法测定丙二醛(MDA)含量。通过电镜观察大鼠卵巢组织超微结构的变化。 结果:经比较更年期针刺组与更年期对照组,更年期针刺组与青年对照组,结果显示:针刺能调节更年期雌性大鼠卵巢组织内NO含量和NOS活性,并且能使SOD活性升高、MDA含量下降;针刺后更年期雌性大鼠卵巢组织超微结构得到明显改善,表现在生长卵泡增多,闭锁卵泡减少,线粒体、滑面内质网及脂滴增加,溶酶体减少。 结论:针刺“足三里”、“关元”穴能增强更年期雌性大鼠卵巢组织抗氧化清除自由基的能力,明显改善卵巢组织的超微结构,调节卵巢生理功能,从而延缓卵巢的衰老进程。

【Abstract】 Purpose: This experiment was to investigate effect of acupuncture at "Zu San Li" and "Guan Yuan" on antioxidation ability and ultrastructure of ovarian organization of menopausal female rats.Method: After acupuncture at "Zu San Li" and "Guan Yuan" of menopausal female aged rats, ovarian albumen content was mensurated with Bradford method;ovarian nitrous oxide (NO) content was mensurated with nitrate reductase method;ovarian nitric oxide synthase (NOS) content was mensurated with spectrophotometry method;ovarian Superoxide dismutase (SOD) energy was mensurated with xanthine oxidase method;ovarian tissue malonaldehyde (MDA) was determined with thiobarbituric acid test;and changing of ovarian ultrastructure was detected with electron microscope.Results: with comparing between the acupuncture group and the comparing model group, it can be conclude that acupuncture at at "Zu San Li" and "Guan Yuan" can adjust NO content and NOS activity of ovaries, increase SOD activity, and decrease MDA content, the ovarian ultrastructure of of the menopausal female rats, consequently, was reformed, which was represented by that there were more follicles, mitochondrial, smooth surfaced endoplasmic reticulum, while less atretic follicles and lysosome.Conclusion: acupuncture at "Zu San Li" and "Guan Yuan" of menopausal female aged rats can improve antioxidation ability to eliminate free radical, modulate ovarian physiological function, and refine ultrastructure of ovarian organization, which will delay ovarian consenescence.
