百令胶囊对糖尿病肾病大鼠保护作用的实验研究

【摘要】 1糖尿病肾病(DN)大鼠模型的建立目的观察模型大鼠一般状况、血压、血糖﹑24h尿蛋白定量、肾功能及病理改变。方法32只大鼠随机分为对照组和DN模型组(模型组),分别按不同观察时间(4、12 w)再分为2组,每组8只。应用链脲佐菌素(STZ)诱导右肾切除大鼠DN模型,测定血糖及24h尿蛋白定量;通过全自动生化仪检测血肌酐(Scr)、尿素氮(BUN),并计算内生肌酐清除率(Ccr);通过光镜观察肾脏病理变化。结果模型组大鼠于注射STZ后2、4、6、8、12w末,血糖明显高于对照组。12w模型组大鼠的平均动脉压、24h尿蛋白定量、BUN、Scr均明显高于对照组大鼠,而Ccr低于对照组大鼠,均P<0.05。结论DN模型制作成功,为探讨药物干预治疗作用及其机制奠定了良好的模型基础。2百令胶囊延缓糖尿病肾病大鼠肾损害的作用研究目的观察百令胶囊对DN大鼠血糖﹑24h尿蛋白定量、肾功能、大鼠肾组织病理变化及转化生长因子-β1(TGF-β1)的影响。方法24只大鼠随机分为对照组、模型组、治疗组,每组各8只。复制模型成功两天后,治疗组每日灌胃百令胶囊(2.5g/kg),模型组及对照组每日给予等量的饮用水,测定血糖及24h尿蛋白定量;通过全自动生化分析仪检测血肌酐(Scr)、尿素氮(BUN),并计算内生肌酐清除率(Ccr);通过光镜观察肾脏病理变化;免疫组化的方法检测肾组织中TGF-β1的表达。结果给药12w后,模型组大鼠血糖、24h尿蛋白定量、Scr、BUN及左肾/体重均明显高于对照组大鼠,而Ccr显著低于对照组大鼠,均P<0.05,且肾组织出现了较明显的病理损伤;对照组大鼠肾组织可低水平表达TGF-β1,模型组大鼠肾组织TGF-β1表达显著上调,均P<0.05。经百令胶囊治疗后,大鼠24h尿蛋白定量、Scr、BUN及左肾/体重均明显减低,均P<0.05,血糖减低,Ccr显著增高,P<0.05,肾组织病理损伤明显减轻,大鼠肾组织的TGF-β1表达显著下调,均P<0.05。结论百令胶囊可通过减少DN大鼠肾组织TGF-β1的表达,而减轻DN大鼠肾组织损伤。故百令胶囊对DN大鼠具有肾保护作用。

【Abstract】 1 Experimental study of rats with diabetic nephropathyObjective To establish model of diabetic nephropathic rats,and to observe their general condition, blood pressure, blood sugar, urinary protein/24h, renal function, pathological changes of renal tissue.Methods 32 rats were randomly divided into control group (control group), diabetic nephropathic model group (model group), which had two sub-groups according to different time (4w, 12w), every group had 8 rats. We established the rat model of diabetic nephropathy induced by streptozotocin (STZ), observed the blood sugar, average arterial pressure, urinary protein/24h, serum creatinine (Scr), blood urea nitrogen (BUN), and counted endogenous creatinine clearance rate (Ccr); we also observed the renal pathological changes by optics microscope and electronic microscope.Results The blood sugar of model group rats which were injected STZ was higher than that of control group in 2w, 4w, 6w, 8w, 12w. Average arterial pressure, urinary protein/24h, SCr, BUN of model group rats were higher distinctly than those of control group, while Ccr of model group rats was lower than that of control group,(respectively,P<0.05).Conclusion We established successfully the rat model of diabetic nephropathy, which was the basis of our futher research of drug intervention.2 Experimental study on effect of Bailin capsule on diabetic nephropathy in ratsObjective To observe the effect of Bailin capsule on blood sugar, urinary protein/24h,renal function,and pathological changes of renal tissue, and the expression of transforming growth factor-β1(TGF-β1) in rats with diabetic nephropathy.Methods 24 rats were randomly grouped into control, model and treatment,8 rats for each group. Two days after models’ establishment,Bailin capsule was administrated (2.5g/kg) by a stomach tube in treatment group, and same amount of water was fed in model and control group. Then we detected the blood sugar, urinary protein/24h, Scr, BUN, calculated Ccr, and used optics microscope to observe the the renal pathological changes. The expressions of TGF-β1 in renal tissue were investigated by immuno-histochemiccl method.Results After 12w, the blood sugar, urinary protein/24h, Scr and BUN in model group were significantly higher compared with control group(respectively, P<0.05). However, Ccr in model group was lower than that of control group, and there was obvious renal tissue damage (respectively, P<0.05). After treatment of Bailin capsule, urinary protein/24h, Scr,
