党参质量控制研究

【摘要】 党参为常用中药,具有化学成分复杂、主要成分含量低、对照品获取困难等问题。现版药典主要限于定性鉴别,定量上仅规定了浸出物含量,不能有效控制其质量。为此,本论文对山西道地药材潞党参进行了系统的质量控制研究,以期建立完善、可控的党参药材质量标准。 主要内容包括有效性和安全性控制两方面。在有效性控制上: 1.建立了潞党参中苍术内酯Ⅲ的高效液相外标测定法,测定方法专一、稳定、快捷。规定了潞党参中苍术内酯Ⅲ的含量限度。 2.首次建立了潞党参石油醚提取物和石油醚提后甲醇提取物的HPLC指纹图谱,并与方山野党、甘肃陇西党参进行了指纹比较。石油醚提取物检测波长为230nm,得到9个共有指纹峰。石油醚提后甲醇提取物方法检测波长为266nm,得到13个共有峰。方法学考察表明,该法的稳定性、重现性均符合指纹图谱分析技术要求,用中药色谱指纹图谱相似度评价系统软件分析结果显示:10批潞党参相似度>0.8,说明此产地药材有比较好的一致性。方山野党、甘肃陇西党参与潞党参相似度<0.5,说明野党与栽培品;不同产地党参之间存在差异。该指纹图谱能够更全面、有效地反应不同品种党参药材的内在质量。 3.补充了党参品质评价常规指标,如水分、总灰分、酸不溶性灰分、多糖、浸出物等项。 4.采用火焰原子吸收分光光度法测定了不同生长年限党参地上部分和地下部分无机有益元素的含量:党参及其栽培土壤样中无机元素的含量。通过测定表明,元素Fe、Zn、Ba、Sr、Na、Ca、Mg地上部分的含量均大于地下部分,其中地上部分和地下部分的Fe、Zn元素随着生长年限的增长均呈下降趋势。栽培土样中元素的含量均大于药材中的含量。计算了党参对Fe、Sr、Na、Ca、Mg的富集系数,其中对Sr的富集能力最强,对铁的富集能力最弱。初步探讨了党参补气益血、治疗心脑血管疾病药理作用与其相应微量元素含量之间的关系。 在安全性控制方面有:

【Abstract】 Radix Codonopsis is a medicinal herb most in use,which has many kinds of components.But the content of main components are low,standard substance is difficult to get.The 2005 Chinese phamacopoeia specifies determination of extractives only. The quality is difficult to control. My experiment studies on the quality control of traditional radix codonopsis plant in shanxi province, in order to establish a perfect quality standard for radix codonopsis.This paper covers validity and security .In validity:1. A method for determination of atractylenoideⅢ in radix codonopsis by HPLC was established. This method is specific、 stable and rapid and it can be used to control the quality of radix codonopsis.The limit content of atractylenoidelll in radix codonopsis was defined.2. A method of the HPLC fingerprint for determination the liposoluble and water-soluble components in radix codonopsis was established.The liposoluble components detection wavelength was set at 230 run, nine peaks were obtained by the HPLC liposoluble fingerprints analysis. Water-soluble detection wavelength was set at 266 nm, thirteen peaks were obtained. This HPLC method has good stability and ruggedness and agree with the demand of fingerprint. The spectra were compared with a software "similarity evaluation system for chromatographic fingerprint of TCM" published by GPC(version 2004A). The results show that the 10 kinds of radix codonopsis with a high similarity(80%).This fingerprint can overall effectiveness reflect the quality of Radix Codonopsis.3. The routine index of quality evaluation of radix codonopsis was measured,eg moisture content, total ash, acid not dissolve ash, polysaccharide ,extractives etc.4. Beneficial and inorganic elements of radix codonopsis was detected in different years and different parts by flame atomic absorption spectrophotometry.It was found that the content of Fe、 Zn、 Ba、 Sr、 Na、
