山茱萸药材及其成药中马钱苷的考察和忍冬植物中马钱苷分布情况研究

【摘要】 第一部分 山茱萸(Fructus corni)为山茱萸科植物山茱萸(Cornus officinalis Sieb. et Zucc.)的干燥成熟果肉,是一种常用的补益中药,主产于浙江、河南、陕西等地。马钱苷是山茱萸中主要的环烯醚萜类成分,文献研究表明其有免疫双向调节作用,并可抗失血性休克。中国药典2005年版将山茱萸中马钱苷含量作为山茱萸药材质量控制指标之一。为了进一步深入研究山茱萸中马钱苷的资源状况,我们通过大孔吸附树脂富集和硅胶柱层析,从山茱萸中分离出马钱苷,同时得到另外两种化合物,利用UV、IR、NMR等技术鉴定了其结构,分别为环烯醚萜类化合物7β-O-甲基莫诺苷和三萜类化合物熊果酸。我们比较了不同产地山茱萸药材及不同生长期山茱萸植物各个部位的马钱苷含量,并以马钱苷为对照,通过对11批山茱萸药材的考察,使用计算机相似度评价软件进行了相似度计算,建立了山茱萸药材水溶性成分的指纹图谱,可有效鉴别山茱萸药材的真伪,为全面评价及控制山茱萸内在质量提供新的方法和理论依据。 明目地黄丸和耳聋左慈丸是以山茱萸为臣药的两种复方成药,均被2005年版药典收载,我们测定了这两种中成药中马钱苷的含量,为更有效的控制其质量提供实验依据。 第二部分 忍冬科植物忍冬(Lonicera japonica Thunb.),别名:金银花、金银藤、二花、二苞花、通灵草,主要产于河南、山东、浙江、四川、陕西、山东、广西、湖南。文献报道,忍冬植物中亦含有马钱苷,为了扩大马钱苷的原料来源和忍冬植物的药用范围,我们通过HPLC法,比较了忍冬植物不同部位中马钱苷的含量,并且测定了不同产地忍冬藤及金银花中的马钱苷含量。更多还原

【Abstract】 Part oneFructus corni is dry ripe sarcocarp of Cornus officinalis Sieb.et Zucc.,family Cornaceae. It is one of beneficial traditional Chinese medicine which can be used for invigorating the liver and kidney ,astringing the preserve essence. The medicinal material is mainly produced in Zhejiang, Henan and Shaanxi. Study on the literatures of iridorid glucosides from Fructus corni show that loganin has immunomodulation effect and anti-hemorrhagic shock activity. Loganin determionation in Fructus corni has been include in China PH. Loganin was got by using macroporous resin and classic preparative method. Meanwhile, I also got another components from Fructus corni. and authenticated their structures by the means of spectroscophy(UV、 IR、 ¹³C-NMR、 ¹H-NMR、 HMBC、 HMQC) .They were 7β-O-methylmorroniside and ursolic acid.We compared the content of loganin in different part and different growing period of Cornus officinalis by HPLC. Besides, through comparing eleven batch of medical materials, we got the fingerprint of the water-soluble components in Fructus corni. By establishing the fingerprint on water-soluble component of Fructus corni, the genuine or fake from the medicinal material was differentiated. In a word, a new method controlling the quality of Fructus corni was established.We also determined the content of loganin in two patent medicine Mingmudihuang pills and Erlongzuoci pills which contained Fructus corni.Part twoLonicera japonica Thunb. belongs to Caprifoliaceae. It was also namedJinyinhua, which is mainly produced in Henan, Shandong and Shaanxi. Study shows that loganin was got from Lonicera. We compared the content of loganin in different part of Lonicera japonica Thunb. by HPLC, and determined the content of loganin in FlosLonicerae and Caulis Lonicerae from different places.更多还原
