低密度脂蛋白对人外周血单核细胞膜联蛋白A1表达水平的影响

【摘要】 目的 探讨低密度脂蛋白(LDL)对人外周血单核细胞(PBMs)中抗炎蛋白膜联蛋白A1(Annexin A1)表达水平的影响,为LDL致动脉粥样硬化机制提供新的视点。 方法 在体外分离培养人PBMs,分别用不同浓度LDL(25mg/L、50mg/L、100mg/L、150mg/L)诱导24小时和用LDL150mg/L诱导不同时间(0h、2h、8h、16h、24h)后,应用流式细胞仪(FCM)检测单核细胞Annexin A1表达水平。 结果 低密度脂蛋白可使单核细胞Annexin A1表达下降((?)<0.05),呈浓度和时间依赖方式。 结论 低密度脂蛋白可能通过降低单核细胞Annexin A1表达而影响单核细胞与内皮细胞黏附,进而启动动脉粥样硬化的发生发展,具体机制有待进一步阐明。
【Abstract】 Objective To explore the effect of low density lipoprotein (LDL) on the expression of Annexin Al in human peripheral blood monocytes (PBMs).Methords PBMs were obtained from the whole peripheral blood drawn from healthy volunteers by centrifugation over Percoll and cultured in vitro. PBMs were stimulated by different concentrations of LDL (25 mg/L, 50 mg/L, 100 mg/L and 150 mg/L) for 12 h and LDL(150mg/L) for 0 h, 2 h, 8 h, 16 h and 24 h. The expressions of Annexin A1 were measured by flow cytometry.Results LDL inhibited the expressions of Annexin A1 in human PBMs as dose- and time-dependent manners(P<0.05).Conclusions LDL may play an important role in the inflammatory and atherosclerotic processes by inhibiting the expression of Annexin A1, which may provide a new insight into the development of atherosclerosis.
