生物组织散射相函数的实验研究

【摘要】 对生物组织的光学参数进行比较准确的测量,是生物组织光学研究的一个重要课题,是进一步研究光在生物组织中传播的基础,对激光外科、光动力疗法等激光临床应用有重要的指导意义。本论文以测量组织散射相函数为主要内容,首先从理论上介绍了当前研究光在生物组织内传输分布的输运理论,介绍了不同偏振方向的光在生物组织体内的传输方程;对当前一些测量散射相函数的文献作了归纳总结,分析了在测量中存在的光源偏振性的问题;从Mie氏理论出发,推导出了不同粒径的球形粒子的散射相函数;计算了不同方向的偏振光入射到不同直径的微球上时,散射光强的比值,得出结论在做测量散射相函数的实验时,为了获得精确的散射相函数,一定要注意光源的偏振性。本文对测量散射相函数的实验作了详细的描述,包括具体的实验装置、实验样品的制备、光源与探测器的选择、实验的测量原理、数据的处理方法,在这里面既有前人的工作,又有我们对其进行的改进。我们的改进工作包括:明确提出了在测量散射相函数时应当选用自然光作为光源,对影响测量HG相函数中的关键量g因子的测量时步长值作了估算,并且通过实验的方法加以验证。本文最后对组织模拟液Intralipid的散射相函数进行了测量,并与用HG相函数表示的Intralipid的散射相函数作了比较,表明HG相函数表示的Intralipid的散射相函数与实际的散射相函数有一定的差别。我们所做的这些工作将为以后进一步研究光在生物组织中的传输提供良好的基础。
【Abstract】 Accurate measurement of tissue optical parameters is very important to tissueoptics.It’s also a basis for further research of light propagation within tissues, whichcan offer vital guidance to laser’s clinic application such as laser surgery and laserdynamic. Scattering phase function is one of the important optical parameters,At thefirst ,The paper introduce the theory of transportation which is used to study thedistribution of scattrring light and transportation in tissue , introduce the transportingequation of different polarized light;The method of the measurement of scatteringphase function in literature are sumerized ,the paper also analysize the problem ofpolarization which has the important relation to the measurement of scattering phasefunction;We can get the scattering phase function of different diameter spheres bythe theory of Mie;Mie theory can also be used to calculate the distribution of thedifferent polarized light which is scattered by the biological tissue particle .It candraw the conclusion that there’s more important influence to measure the biologicaltissue phase function with different polarized light.So,to get accurate phase functionby experiment ,the natural(unpolarized)light source should be choosen.The paper describe the experiment in detail,including the device of the experiment、the preparation of sample、the principle of measurement、the method of how to get thedate that we need.There are a lot of work of other people in the paper and we alsogive new advice to the experiment. The first is that it must be used the unpolarizedlight source when measuring the scattering phase function .The second we calculatethe influence of different step length when measuring g . g is the important parameterof HG phase function. The experiment prove that our estimate is correct.At last wemeasure the scattering phase function of Intralipid which is a kind of simulativesolution of biological tissue and draw the conclusion that there is more differentionbetween the HG phase function and the measured phase function by comparingthe HG scattering phase function with the measured scattering phase function. Whatwe have done will provide a favorable basis for further research.
