雷公藤内酯醇与传统抗肿瘤药联合抗人肝癌SMMC-7721细胞活性研究及机理探讨

【摘要】 雷公藤内酯醇与传统抗肿瘤药物阿霉素和羟基喜树碱联合作用对体外培养人肝癌SMMC-7721细胞,主要检测指标是细胞形态观察,台盼蓝染色,MTT染色,DNA ladder等,检测药物对肿瘤细胞的杀伤作用。 SD大鼠经济、易饲养、易获得,且模型成功率高,自然消退率低,是建立移植性肝癌模型的理想选择。将人肝癌SMMC-7721细胞皮下种植在SD大鼠上,静脉注射雷公藤内酯醇,游标卡尺测量肿瘤体积大小,软件分析数值可靠性。 Caspase家族属于天冬氨酸特异的半胱氨酸蛋白酶,围绕活性中心位点半胱氨酸的氨基酸残基是保守的。正常情况下,胞质中的Caspase-3以无活性的酶原形式存在,只有当细胞凋亡时,才被激活为有活性的Caspase-3。测定肿瘤细胞培养时药物联合作用细胞时caspase-3表达情况,以探讨药物联合作用的分子生物学机理。 与对照组相比,体外培养肿瘤细胞在用甲素处理后活细胞大幅减少,而甲素与阿霉素或羟基喜树碱联合作用比单独用一种药物的杀细胞效果要强得多。体内接种肿瘤细胞在用药后,肿瘤块体积较小且增长缓慢,而对照组肿瘤体积却大幅增长;雷公藤内酯醇与阿霉素或羟基喜树碱联合用药作用体外培养肿瘤细胞后,caspase-3表达明显上调。 得出结论:雷公藤内酯醇对体外和体内培养的人肝癌SMMC-7721细胞有明显的促凋亡作用。它与传统抗肿瘤药联合作用该细胞时呈现强烈的协同作用;其对肿瘤细胞的杀伤作用与caspase-3的激活有关。
【Abstract】 With the co-action of Triptolide with two traditional anti-tumor medicines Amycin orHydroxycamptothecine on human heptocarcinoma SMMC-7721 cell, death-inducingeffect of triptolide’co-action with these two traditional anti-tumor medicines wasevaluated. The vitro culture examination indexes included observation of cellappearance, trypan blue staining, MTT staining and DNA laddering.SD rats are easy to obstain. It is easy and economic to feed SD rats, most important ofall, the rate of tumor’s survival is very high, while the rate of tumor’ fadeaway is verylow. SD rats are ideal model to act as transplanted liver cancers. In this essay,afterinoculation of SMMC-7721 to SD rats, the way of weighing the effect of triptolide onSD rats’ death was through the examination of the mass of tumor by the verniercaliper, and software was available to value the reliability of the number.Caspase family belongs to cysteine aspartate-specific protease. Normally caspase-3exists in the cytoplasm with the activity-free zymogen. When cells neoresis, thezymogen is activated. We can measure the expression of caspase-3 with theexpression of caspase-3 to investigate the molecular mechanism of this coaction ofthese chemicals.Compared with the control group, the living cells were reduced significantly after theapplication of triptolide’s cooperation with two traditional anti-tumor medicines, thetumor’s volume were smaller and increased slowly while the control group’s tumorvolumes increased significantly. At the same time, the expression of caspase-3 risedsharply.As a result, a conclusion could be drawn: Triptolide displayed a co-action effect whencooperated with the two traditional medicines, and triptolide had obvious lifeinhibition and apoptosis induction both in vivo and in vitro, which was associatedwith the expression of caspase-3.
