蛹虫草培养基的综合利用研究

【摘要】 本研究课题深入而系统地研究了蛹虫草固体培养基中虫草素虫草多糖的提取纯化工艺、检测方法、结构鉴定和虫草多糖抗氧化活性以及蛹虫草的固体液体发酵,得到了满意的结果。 1.对蛹虫草进行固体液体发酵,应用高效液相色谱对液体发酵的菌丝和发酵液中虫草素的含量进行检测,结果发现菌丝在第七天,发酵液在第八天虫草素的含量最高。固体发酵确定40天为一个周期,蛹虫草的长势良好。 2.对影响虫草素提取率的工艺参数进行正交实验研究,确定用75%乙醇超声波提取三次,40min/次,料液比为1:20的工艺为最佳提取工艺。 3.对影响虫草多糖提取率的工艺参数进行正交实验研究,确定用水80℃提取三次,90min/次,料液比为1:20的工艺为最佳提取工艺。 4.从优化的虫草素和虫草多糖生产工艺出发,研究出同时提取虫草素和虫草多糖的工艺方法。同时,分别采用高效液相和苯酚-硫酸法对虫草素和虫草多糖进行含量测定,确定不同纯化方法所得虫草素虫草多糖的含量差异,进一步确定分别使用阴离子树脂和DEAE纤维素为最好的纯化方法。 5.RP-HPLC对不同产地人工蛹虫草子实体中虫草素的含量进行了测定,发现不同产地的人工蛹虫草中虫草素的含量差异很大,其中以泾阳虫草为最高,江苏七庙2号次之。同时,同一种蛹虫草中虫草素的含量子实体多于培养基。 6.应用熔点测定、IR、以及元素分析对所得的结晶Ⅱ进行结构鉴定,结果与虫草素的资料数据一致。 7.研究了15%、30%、50%、70%四个不同醇沉部分虫草多糖的抗氧化活性,结果发现四部分虫草多糖都有一定的抗氧化性,其中30%部分抗氧化活性最强。
【Abstract】 The research on extraction and purification technique, examination method,construction identification of cordycepin and exopolysaccharide in the solid of Cordyceps Militarism Link,as well as exopolysaccharide antioxidation activity and the two fermentatin methods, had been done indepthly and systematically, the result are satisfactory.1. Through study on solid and liquid ferment we found that the highest content of cordycepin is seventh and eighth day in mycelia and liquid culture media respectively by HPLC in liquid ferment; in solid ferment forty days as a periods is god for mycelia growth.2. The optimal technique parameter of extracting cordycepin is researched by orthogonal design. The result is that the best extractive condition of cordycepin was use ultrasonic extraction three times in twenty times of 75% ethanol solution,and 40 mins each time.3. The optimal technique parameter of extracting exopolysaccharide is researched by orthogonal design. The result that The best extractive condition of exopolysaccharide was extracted three times in twenty times of water,and 90 mins each time.4. According to the optimal technique of extracting cordycepin and exopolysaccharide. The studies on the method of extraction cordycepin and exopolysaccharide at the same time had done. Respectively.cordycepin and exopolysaccharide have been determined by HPLC and phenol-sulfuric acid method ; it is the different content in cordycepin and exopolysaccharide through the different purification methods.5. In the different artificial Cordyceps Militarism Link, the content of cordycepin is different and obvious by the measurement of RP-HPLC, the highest is shannxi jingyang Cordyceps Militarism, xiuzhou qimiao No.2 is secondly, the content of cordycepin is more high in mycelia than in the culture media in the same kind of Cordyceps Militarism.6. The crystallize II have been characterized by TLC,HPLC, Melting Point Determine,IR, and chemical element analysis, the result is in accordance with the cordycepin spectrum data.7. Studying 15%,30%,50%,70% four different ethanol density exopolysaccharide antioxidize activity.the result showed that all of the four-part have antioxidize activity .but the 30% parts is the most strong.
