一株桃耳七内生菌产鬼臼类化合物的研究

【摘要】 鬼臼毒素是一种具有抗肿瘤和杀虫活性的木脂素类化合物,不仅是治疗尖锐湿疣的良好药物,亦是合成多种治疗癌症药物的前体物质,具有很大的药用价值。因而人们对鬼臼毒素的需求量日益增加,但其天然资源十分有限,故如何合理地利用并开发其资源就成为人们研究的一个热点。 本研究的目的:获得可以利用微生物发酵产生鬼臼毒素的内生菌,为扩大鬼臼类物质资源开辟一条新的途径。采用微生物分离方法,分离筛选出一株真菌——Iml;对其形态和生长特征进行研究,初步鉴定该菌株属于半知菌亚门,丛梗孢目,暗梗孢科,交链孢霉属(Alernaria Nees ex Wall.)。对Iml分批发酵培养,发酵产物依次用氯仿、正丁醇提取,硅胶柱层析,制备型薄层层析以及重结晶等方法分离纯化,获得了三种化合物。经薄层层析(TLC),高效液相色谱(HPLC),碳、氢核磁共振谱分析,得出:化合物Ⅰ为鬼臼毒素,化合物Ⅱ为山奈酚,化合物Ⅲ初步认定为鬼臼毒素单糖甙类化合物。Iml具有良好的遗传稳定性,在PDA液体培养基中生长周期为7d-9d。PH 6.0~6.5,温度25℃,是其发酵产鬼臼毒素的有利条件。 研究证明利用药用植物内生微生物发酵生产药用植物活性物质是可行的。为植物原料药资源开发开辟了新途径。对保护生态环境具有重要的意义。

【Abstract】 Podophyllotoxin is a kind of anticancer and insecticidal lignans. It is an effective medicine in treating wart and a precursor of some anti-tumor medicine. Because of its great medical potential, podophyllotoxin has been demanded desperately. However, the natural resource of it is very limited. Therefore, how to utilize and develop its resource reasonably is a hot point.The experiment tries to obtain podophyllotoxin through microbial fermentation to settle the problem of resource shortage. An endophytic fungi Iml was isolated from Sinopodophyllum emodi (Wall.) Ying . The fermentation production was extracted by chloroform and /rbutanol and the extraction was collected respectively. Three compounds were separated from two kinds of extract through silica gel C. C. (200-300 mesh) , TLC and recrystallization. Compound I was identified as podophyllotoxin and compound II as kaempferol, compound III was initially considered as a kind of podophyllotoxin- 0- Glu type on the basis of TLC, HPLC and 1H, 13C-NMR. The experiment also studied the biological character of the strain Iml. The initial result shows that Iml belongs to Fungi Imperfecti, Moniliales, Dematiaceae, Alternaria Nees ex Wall. Iml grows well in PDA medium and has a good genetic stability. The optional fermentation condition of Iml was also investigated: PH 6.0—6.5, temperature 25°C.The result fully indicates that it is feasible to use endophytic microbial of anti-cancer medicinal plant to produce anticancer substance. The research pioneer a new gateway for the development of medicine resource, and it has a very important meaning to protect biological environment.
