注射用黄芪冻干粉对大鼠心肌缺血的保护作用及分子机制

Study of Protective Effect of Lyophilized Powder of Astragalus Injection of Myocardial Ischemia in Rats and Molecular Mechanisms

【摘要】 目的 观察注射用黄芪冻干粉对心肌缺血的保护作用,并对其可能的作用机制进行初步探讨。 方法 建立心肌缺血模型,使用BL-420生物信号采集与处理系统记录大鼠血流动力学指标,HE染色观察正常和模型损伤以及用药组心肌的超微结构,Low Shear-30血液流变仪检测血液流变学指标,分光光度计测量各组MDA,LDH,GSH-Px含量,放射免疫法检测血浆内皮素含量,免疫组织化学方法检测各组左室心肌Bax,Bcl-2蛋白表达。 结果 血流动力学指标:各组在结扎后心率、LVSP,dp/dmax,均明显降低,-dp/dmax、LVEDP明显升高,与模型组比,注射用黄芪冻干粉能有效回升心肌缺血大鼠HR、LVSP、dp/dtmax、有效降低心肌缺血大鼠LVEDP、-dp/dtmax。

【Abstract】 Objective To study the protective effects of Lyophilized Powder of Astragalus injection on Hemodynamic, Hemorheology, endothelin, apoptosis, lipid peroxidation and immunochemistry aspects to get further understanding of its mechanism.Methods Randomly allocate SD rats into groups to form Myocardial Ischemia models, left anterior descending branch of coronary arteries of SD rats were Ligated to observe the change of hemodynamics by using BL-420 biological signal collecting and disposal system, the ultrastructure observation of Myocardial HE pigmentation, Low Shear 30 Hemorheology instrument to test blood Hemorheology, The level of lactate dehydrogenase(LDH)and malondialdehyde(MDA),GSH-Px in plasma were measured by spectrophotometer, the contents of endothelin(ET) in plasma was determined by radioimmunology. The changes in the expression of Bcl-2 and Bax proteins in left ventricular myocardium were observed by immunohistochemical method.Results Hemodynamics: Lyophilized Powder of Astragalus injectioncan significantly protect cardiac muscle of the acute ischemia by increasing HR, LVSP, dp/dtmax while falling LVEDP, -dp/dtmax, compared with the model. The contents of endothelin(ET) in plasma: ET was obviously increased in the model, as dropped after the administration of LPA injection and marked of large dosage. Lipid peroxidation index: The level of LDH and MDA were increased in model groups and after dose Lyophilized Powder of Astragalus injection, they dropped with statistical meaning. Morphology: The normal Myocardial cells have the natural conformation with the intact structure. The membrane of ischemic Myocardial cells is complete as well as the apparently damage of the cell structures. The expression of Bcl-2 and Bax proteins in left ventricular myocardium: Lyophilized Powder of Astragalus injection groups could obviously decrease the protein expression of Bax gene, but increase the protein expression of Bcl-2 gene. Hemorheology: Lyophilized Powder of Astragalus injection can reduce the whole blood high-, mid-and low-shear viscosity as well as hematocrit after their increase by ligating SD rats’ left anterior descending branch of coronary arteries.ConclusionsLyophilized Powder of Astragalus injection can protect ischemicMyocardial, the mechanism of which possibly consist of its multifunctional effects, including improving the systole and diastolic function by ameliorating the disorder of hemodynamics of Myocardial Ischemia, adjusting the Hemorheology, decreasing the contents of endothelin in plasma, dropping LDH, MDA to inhibit the lipid peroxidation and reducing Myocardial cell apoptosis.
