藏猪氟烷基因、雌激素受体基因和生长激素基因PCR-RFLP分析

【摘要】 本实验以藏猪作为研究对象,采用PCR-RFLPs方法分析了藏猪94个个体氟烷基因HhaⅠ、88个个体雌激素受体基因PvuⅡ和94个个体生长激素基因ApaⅠ、PvuⅡ、BstⅪ和HaeⅡ酶切位点多态性。 分析结果表明:氟烷基因全部为NN基因型,不存在氟烷隐性基因纯合子和杂合子个体,说明藏猪抗应激能力强。雌激素受体基因AA基因型频率为11.36%,AB基因型频率为64.77%,BB基因型频率为23.86%;A、B两种等位基因频率分别为43.75%和56.25%;B等位基因频率显著高于A等位基因频率(P<0.05);B等位基因是产仔数增效基因,B等位基因频率较高说明藏猪具有较高的繁殖潜能。 生长激素基因ApaⅠ酶切位点有A、C两种等位基因,AA、AC和CC三种基因型;AA基因型频率为7.5%,AC基因型频率为24.5%,CC基因型频率为68%;等位基因A的频率为19.8%,等位基因C的频率为80.2%,等位基因C的频率显著高于等位基因A的频率(P<0.05)。BstⅪ酶切位点有A、B两种等位基因,AA、AB和BB三种基因型;AA基因型频率为2.1%,BB基因型频率为86.2%,AB基因型频率为11.7%;等位基因A的频率为8.0%,等位基因B的频率为92%,等位基因B的频率极显著高于等位基因A的频率(P<0.01)。PvuⅡ酶切位点有A、B两种等位基因,AA、AB和BB三种基因型;AA基因型频率为6.4%,AB基因型频率为2.1%,BB基因型频率为91.5%;等位基因A的频率为7.5%,等位基因B的频率为92.5%,等位基因B的频率极显著高于等位基因A的频率(P<0.01)。HaeⅡ酶切位点有A、B、C三种等位基因,检测到AA、BB和BC三种基因型;AA基因型频率为2.1%,BB基因型频率为75.5%,BC基因型频率为22.4%;等位基因A、B和C频率分别为0.145、0.745和0.110,差异水平达到极显著(P<0.01)。

【Abstract】 In this thesis, HhaI polymorphisms of halothane gene in 94 Tibetan pigs and Pvu II polymorphisms of ESR gene in 88 samples and Apa I 、 Pvu II 、 BstXI、 Hae II polymorphisms of growth hormone gene in 94 Tibetan pigs were detected by PCR-RFLPs.The results of polymorphisms of restriction enzyme sites were indicated as follow: the genotypes of halothane were NN. This showed that Tibetan pig had the strong capability of resisting stress. The frequency of AA genotype of the ESR gene was 11.36%. AB was 64.77%. BB was 23.86%. The frequency of A allele gene was 43.75% and B was 56.25%. There was a significant difference between A and B allele gene (P<0.05).B allele was a dominating gene of the porcine number born. The high frequency of B allele indicated that Tibetan pig had high potential competency of reproduction.After the growth hormone gene was digested by Apa I, two alleles and three genotypes (AA, AC and CC) were detected. The frequency of AA, AC and CC genotypes were respectively 7.5%, 24.5%, 68%. The frequency of A allele gene was 19.8% and C was 80.2%. There was a significant difference between A and C allele gene (P<0.05). BstXI digestion revealed two alleles and three genotypes (AA, AB and BB). The frequency of AA, AB and BB genotypes were respectively 2.1%, 11.7%,86.2%.The frequency of A allele was 8.0% and B was 92%.The frequency of A and B was a signification difference (P<0.01) . PvuII digestion revealed two alleles and three genotypes (AA, AB and BB). The frequency of AA, AB and BB genotypes were respectively 6.4%, 7.5%,92.5%.The frequency of A allele was 7.5% and B was 92.5%.The frequency of A and B was a signification difference (P<0.01) . Three alleles and three genotypes (AA, BC and BB) were shown after digestion with Hae II. The frequencies of the three genotypes were respectively 2.1%, 22.4%, 75.5%. The frequency of A allele wasl4.5 %, B was 74.5% and C was 11%. There was a very signification difference among three alleles (P<0.01).
