GTPase在肝癌细胞系中的表达和意义

【摘要】 为了研究GTPase在肝癌细胞系中的表达,探讨GTPase在肝癌发生发展中的意义,运用Western blot测定了GTPase 3个代表性蛋白Racl、Cdc42、RhoA的蛋白表达水平。结果发现,与正常肝细胞系对照,肝癌细胞系中Racl、Cdc42、RhoA蛋白表达水平均上调,提示这3种蛋白可能在肝癌的发生发展中起着重要的作用。同时还发现肿瘤高转移细胞株中的Racl、Cdc42、RhoA的蛋白水平显著高于其在肿瘤低转移细胞株中的表达,提示Racl、Cdc42、RhoA可能与肿瘤的转移有密切关系。 在进一步的研究中,选择Racl为代表蛋白,用特异性针对Racl的siRNA干扰肿瘤高转移细胞株97-H。结果发现,RNA干扰后Racl蛋白在肿瘤高转移细胞株97-H中的表达明显减少,97-H细胞的生长、细胞活力及细胞在体外侵袭能力受到明显抑制,证明Racl与肝癌的发生发展及转移有密切的关系。同时也为肿瘤的基因治疗提供了新的靶点和策略。

【Abstract】 The expression of three important proteins of Rho GTPases was studied in hepatoma cell lines and the significance of GTPase in the occurrence and development of hepatoma was also investigated. Protein expression levels of Racl, Cdc42 and RhoA in several hepatoma cell lines were examined by Western-blot. Compared with normal liver cell line, protein expression levels of Racl, Cdc42 and RhoA in three hepatoma cell lines were up-regulated, which implied that Racl, Cdc42 and RhoA might play an important role in the development and progression of hepatoma.In further research, the small interference RNA technique was employed to knock down gene expression of Racl in hepatoma cell line 97-H. The protein level of Racl, the cell proliferation, the cell vitality and the ability to invade the reconstituted basement membrane were decreased dramatically after transfection with a Racl-specific siRNA vector .The results suggested that Racl might play an important role in the development, progression, transference of hepatoma and also provide a possible strategy for the interference of tumor angiogenesis by targeting the Racl.
