复方叶下珠对人肝癌细胞移植裸鼠HBxAg表达的影响及对HCC的防治意义

【摘要】 目的 观察复方叶下珠(Compound phyllanthus urinaria L.,CP)对人肝癌细胞(HepG2.2.15)移植裸鼠(Balb/c-nu mice)HBxAg表达的影响,分析CP对肝细胞肝癌(Hepatocelluar carcinoma,HCC)的防治意义,探讨其对HCC的作用是否与HBxAg表达有关。 方法 将40只Balb/c-nu小鼠随机分为五组(1st,CP预防组;2nd,CP治疗组;3rd,CTX(环磷酰胺)治疗组;4th,模型对照组;5th,空白对照组),制作人肝癌细胞(HepG2.2.15)皮下移植瘤模型,分别采用不同方法给药,观察裸鼠及移植瘤生长情况,实验结束时称量瘤重、用ABBOTT AxSYM系统检测外周血清甲胎蛋白(Alpha fetoprotein,AFP)、免疫组化(Immunohistochemistry,IHC)技术检测乙型肝炎病毒X基因表达产物HBxAg在瘤组织、肝组织中的表达。 结果 1st组造模3周成功率85.7%(6/7),其它造模组为100%(8/8),相互无统计学意义(P>0.05)。不同组裸鼠体重增长与观察时间之间存在交互作用(P<0.05)。1st,2nd,3rd组分别与4th组比较,移植瘤重之间差别有统计学意义(P<0.05),而1st,2nd,3rd各组两两比较无显著差别(P>0.05)。各实验组外周血清AFP含量两两之间比较无统计学意义(P>0.05)。各用药组对肿瘤和肝脏组织切片中HBxAg表达均有抑制作用(P<0.05),其表达程度具有相似性:4th组>2nd组>1st组>3rd组,但是否存在因果关系尚难定论。在瘤组织中,3rd组表达最少,1st组次之:但1st、2nd、3rd组之间无统计学意义(P>0.05)。 结论 1.HBxAg在移植瘤与荷瘤裸鼠肝脏中均有强表达;2.CP不能完全阻断移植瘤的发生,但能减缓移植瘤形成,抑制肿瘤的生长和HBxAg的表达,后者可能是CP防治HCC的主要机制之一;3.裸鼠外周血清AFP含量检测没有明确反映CP与移植瘤的生长的关系;4.提示预防是防治HCC的核心。

【Abstract】 Objective: To observe the effects on HBxAg expressions of nude mice (Balb/c-nu mice) transplanted by human hepatic carcinoma cells (HepG2.2.15 cells) and analyze the significance of Hepatocelluar carcinoma (HCC) prevention & treatment of CP (Compound phyllanthus urinaria L.). To explore whether the mechanism of the effects of CP on HCC is correlated the expressions of HBxAg (Hepatitis B x antigen).Methods: 40 Balb/c-nu mice were divided equably and randomly into five groups(1st, Prevention group with CP; 2nd ,Treatment group with CP; 3rd,Treatment group with CTX; 4th,Model control group; 5th,Blank control group) and supplied by different methods of administrations before or after making hypodermic tumor models in nude mice transplanted by HepG2.2.15 cells. To observe the tumor weight of different groups of nude mice, the content of peripheral serum AFP assayed by AxSYM system, and the expression of HBxAg in liver tissue and tumor tissue assayed by immunohistochemical methods.Results: The rate of success of nude mice transplant tumor models in the 1st group was 85.7%(6/7) which is 100%(8/8) in other model groups. And there’s no statistic significance (P>0.05) between these groups. It existed an ubteraction of mice weight and observation periods among different groups (P<0.05). There was no significant difference of tumor weight between 1st, 2nd, and 3rd group (P>0.05). No significant differences of serum AFP concentration were observed between these groups (P>0.05). In experimental groups, the expression of HBxAg in tissue slices of livers were down regulated (P<0.05) as well as in tissue slices of Tumors. There was a comparability of the expression of HBxAg between tissue slices of livers and tumors: 4th>2nd>1st>3rd. But it was unclear about the relationship between the causes and the effects. In tumor tissues, the expression of the 3rd group was least, which of the 1st one took second place. There was no obvious difference between group l-3(.P>0.05).Conclusions: 1. HBxAg expressions can be observed both in tumor tissues and liver tissues of mice transplanted by human hepatic carcinoma cells. 2. CP can’t stop the occurrence of transplanted tumor while can delay it and repress the growth of tumor. It could also inhibit the expression of HBxAg which is probably an important mechanism associated the efficacy of prevention & treatment of CP on HCC. 3. The relationship between CP and the growth of transplanted tumor can’t be reflected clearly by the detection of AFP concentration in peripheral serum of nude mice. 4.Prophylaxis is probably the sticking point of prevention & treatment of HCC.
