管立舒滴丸改善胸痹证的药效物质及相关药效学研究

【摘要】 中药毛冬青具有活血化瘀,通脉止痛的功效,临床上常用于胸痹证的治疗,并显示了较好的开发前景。但目前还存在着药效物质基础不清、制备工艺落后、质控指标欠合理、药效研究仍需深入等问题。因此,本课题拟对毛冬青药效物质基础及其制剂管立舒滴丸改善胸痹证的相关药效学进行研究,以期进一步阐明毛冬青药效物质基础。 本研究采用垂体后叶素所致大鼠急性心肌缺血模型,以T波的变化率作为筛选指标,简单易行,直观可靠;以药效学为指导,突破了普遍存在的单纯以含量筛选工艺的模式,筛选得到毛冬青有效部位,并对其所含的化学成分进行了分离鉴定,首次从毛冬青分离出木脂素类有效成分——新毛冬青素。以新毛冬青素为质控指标,药效学实验做为佐证,优化并确定了管立舒滴丸的制备工艺,为管立舒滴丸的疗效提供了保证。 在对药效物质研究的基础上,对毛冬青制剂——管立舒滴丸改善胸痹证的相关药效学进行了研究。本文采用垂体后叶素所致大鼠急性心肌缺血的模型,研究对心肌缺血的影响;应用颈动脉温度测定法及胶原蛋白—肾上腺素诱发小鼠体内血栓形成的方法以及对三种诱导剂诱导的血小板聚集率的测定,研究对血栓形成的影响;通过对血浆复钙时间、凝血酶原时间以及纤溶活力的测定,研究管立舒滴丸的抗凝作用;通过测定大鼠血液流变性,研究对血瘀的改善程度。上述药效学实验表明,管立舒滴丸具有活血化瘀的功效,对以血瘀为主的胸痹心痛有一定的治疗作用。 本课题还对管立舒滴丸的急性毒性进行了研究,确定了最大给药量,为临床的安全用药提供了科学依据。 另外,本课题采用栓线法对管立舒滴丸抗脑中风的药理作用作更多还原

【Abstract】 Radix Ilics Pubescentis whose principle effects are promoting blood circulation, relieving blood stasis and relieving pain, is mainly and successfully used to treat coronary heart disease and has good prospect to be developed into new preparation. However, some limitations are still in existence. The medicinal substances, preparation technics, quality-control methods and pharmacological effects are still needed to be researched deeply. Therefore, this project is going to find new medicinal substances and to prove the pharmacological effects of Radix Ilics Pubescentis and its dripping pills, Guanlishu, in order to expound its basic kinds and effects of medicinal substances.The variety-rate of T-wave of experimental acute myocardial ischemia rat is used to evaluate the effects of Radix Ilics Pubescentis in this project, which is simple and reliable. It has been used to evaluate the preparation routes and sieving the medicinal substances. Furthermore, a new compound, which is lignan and was named as Neoilexin, was found and used to control the quality of preparations. With the determination of this new medicinal substance and illustration of pharmacological effects, the preparation route is selected confidently.On the fundamental researches of medicinal substances, the pharmacological effects of Guanlishu dripping pill are been work over更多还原
