补益脾肾法对阿霉素肾病大鼠肾脏ET-1和TGF-β1表达的影响

【摘要】 目的:观察补益脾肾之中药复方对ET-1、TGF-β1在阿霉素肾病大鼠肾组织的表达所产生的影响,探讨补益脾肾中药复方对慢性肾脏疾病治疗作用及阻止肾纤维化的可能机制。 方法:将雄性SD大鼠随机分为空白组、模型组,将模型组采用尾静脉一次注射阿霉素溶液(剂量6.5mg/kg),造模后第14天将模型组按24小时尿蛋白定量随机分为模型对照组、苯那普利组、中药复方低、中、高组。分组的当天开始给药,连续给药四周。造模后第14天、第28天、第42天测定大鼠24h尿蛋白,给药四周末,处死大鼠,测定血肌酐、血尿素氮、尿肌酐。取大鼠左肾,用免疫组化半定量检测各组大鼠肾组织ET-1及TGF-β1表达。 结果:(1)阿霉素肾病(AN)大鼠模型于造模后第7天出现蛋白尿,14天左右尿蛋白排泄达到高峰。(2)补益脾肾中药复方、苯那普利组均能明显降低AN大鼠的24小时尿蛋白量。其中,中药高剂量组作用尤为明显。(3)免疫组化病理显示,阿霉素肾病组鼠肾组织中ET-1、TGF-β1表达明显增加(与空白组比较,P<0.05);各治疗组大鼠肾组织中ET-1及TGF-β1的表达均低于模型组。 结论:补益脾肾法不仅能减少阿霉素诱导的SD大鼠模型24小时尿蛋白排泄量,而且能通过对肾组织内皮素-1和转化生长因子β1表达的负性影响,对肾脏纤维化起到可能的延缓及阻止作用。

【Abstract】 OBJECTIVE: To investigate the effects of Reinforcing Spleen and Kidney on expression of ET-1 and TFG-β1 in kidney of The Adriamycin nephropathy rats, and examine the therapeutic function of the compound Chinese formula on chronic renal diseases, as well as the possible mechanism to block the renal fibrosis of it.METHODS: The Sprague-Dawley rats were randomly assigned into control and normal control groups.The Adriamycin-induced nephropathy model rats were established by single injection of Adriamycin(dosage 6.5mg/kg) on the control group.On the 14th day of accomplishment of the injection,the control group were divided into five groups: module,low-dose Chinese formula, moderate-dose Chinese formula, high-dose Chinese formula, benazepril-treated group.Medication was exerted continuously for four weeks from the 14th day.The level of urine protein was measured at each time point.At the end of the medication for 28 days,all of the rats were killed,the level of serum creatinine,urea nitrogen, and urine creatinine were measured,and the pathologic change of kidney was observed.And the protein expression of ET-1 and TGF-β1 were detected by immunohistochemistry.RESULTS: (1)The proteinuria of control group was observed on the 7th day of accomplishment of the injection of adriamycin.And the summit was
