香菇多糖对骨髓抑制贫血小鼠造血调控的影响

【摘要】 目的 本课题采用实验血液学的方法,探讨香菇多糖对造血的调控作用,为香菇多糖在临床的应用及支持通江老区因地制宜开发香菇资源等提供实验室依据,并对中医血化生模式及营卫同源、营卫交会的机理进行初步理论探索。 方法 1.采用复合造模法形成小鼠骨髓抑制贫皿模型,将香菇多糖按高中低不同剂量分组给药,进行外周血、骨髓血细胞的形态学观察及细胞分类计数,并通过流式细胞仪进行细胞各时相周期分析。 2.采用造血祖细胞培养法,观察香菇多糖体内用药对骨髓抑制贫血小鼠各系造血祖细胞集落产率的影响;观察香菇多糖不同浓度体外给药对各系造血祖细胞集落产率的影响。 3.采用ELLISA法测定香菇多糖不同浓度、不同时间点上清液的IL-6水平,观察香菇多糖对基质细胞分泌IL-6的影响。 结果 结果表明,在一定浓度范围内: 1.香菇多糖可升高骨髓抑制贫血小鼠外周血象的白细胞及红细胞数量,提高骨髓有核细胞数,促进骨髓有核细胞由静止的G0/G1期进入增殖期。 2.香菇多糖在体内、外用药均可有效的提高粒、红系造血祖细胞的集落产率,而对巨核系祖细胞的产率影响似不明显。 3.香菇多糖体外用药对贫血小鼠基质细胞分泌IL-6具有显著的刺激作用,且在一定的浓度范围内具有量效关系。 实验结果为香菇多糖的临床应用提供了有价值的实验依据,同时也为中医血化生模式、营卫同源、营卫交会的理论探讨提供了有价值的资料。
【Abstract】 Objective To examine the effect of LNT (lentinan) on aplastic anemia mice Periphery blood, BMC amount and cell cycle; also evaluate the influence of hemopoietic progenitor cell unit and K562 cultivated in vitro affected by LNT and variety of cytokin IL-6 excreted by bone marrow stromal cell, for Investigating the hematopoietic regulation effect of LNT and study the modern mechanism of Ying-Wei tongyuan, Ying-Wei jiaohui addressed by Chinese traditional medical theory.Methods 1 BalB/c Mice were established AA model and divided into four groups, treated by different concentration LNT, To count the amount of WBC, RBC, Hb, PLT and bone marrow nucleated cells , Observe the cell cycle by FCM (flow cytometry)2. Cultivated BMC extracted from aplastic anemia mice in vitro and account the unit of hemopoietic progenitor cell, and evaluate the different effect changed by different concentration of LNT in vitro.3 Adope enzyme linked immunosorbent assay (ELISA) to investigate the different concentration of cytokine IL-6 excrete by bone marrow stromal cell cultured in vitro affected by different concentration LNT. Results Experimental results indicate: Within the range of certain concentration:1 LNT can increase the amount of WBC, RBC and BMC but not include blood platelet, also stimulate more BMC into cell cycle, promote.
