“芪香”气雾剂毒理安全学及其对小鼠免疫功能影响的实验研究

【摘要】 目的: 研究“芪香”气雾剂的药物稳定性试验和毒理安全学试验,评价其稳定性和安全性。研究“芪香”气雾剂对小鼠的非特异性免疫功能、体液免疫和细胞免疫功能的影响,从细胞和分子水平分析“芪香”气雾剂对机体免疫功能的影响,并初步探讨了“芪香”气雾剂的免疫调节作用。 方法: 1.在37℃和相对湿度75%的环境中加速试验三个月,测样品中黄芪甲苷含量变考察其稳定。采用霍恩氏法测定两种性别大、小鼠急性毒性试验的LD50;采用Ames试验、小鼠精子畸形试验和骨髓微核试验,考察“芪香”气雾剂的致突变作用。 2.昆明种成年小鼠随机分4组:空白对照组,免疫抑制模型组,免疫抑制模型+“芪香”气雾剂组,“芪香”气雾剂组。测定各组小鼠免疫器官重量、小鼠单核巨噬细胞吞噬功能(碳粒廓清法)、小鼠NK细胞活性、小鼠血清溶血素、小鼠外周血T淋巴细胞亚群、小鼠脾细胞诱生的IL-2活性、小鼠腹腔巨噬细胞诱生TNF-α含量,并对各组进行分析比较。 结果: 1.“芪香”气雾剂对两种性别大、小鼠急性毒性,LD50均大于

【Abstract】 Objective:The stability and toxicology of "Qixiang" aerosol was studied to evaluate its stability and safety. To preliminary study the effect of "Qixiang" aerosol immunological function in mice. Methods:1. To inspect stability of sample, the contents of astragalin was tested by Accelerating experiment. To inspect acute toxicity by mice LD50 and effect of mutagenesis by Ames test, sperm shape abnormality test in mice and bone marrow cell test.2. Mice were randomly divided into normal control group, Cy group(Using the model of immune function in low level induced by cyclophosphamide), Cy +"Qixiang" aerosol group, "Qixiang" aerosol group. To Measure the index of Thymus and Spleen in mice, the phagocytic index of mononuclear macrophage(carbon granular clearance test), the Cytotoxic activity of splenic NK cell in mice, the half number hemolysis rate in serum mice(HC50), T-lymphocyte subsets in peripheral blood, the contents of cytokine IL-2 induced by splenic cell, the contentsof TNF-a induced by peritoneal macrophage. Compared and analyzed each group.Result:1. Acute toxicity test showed LD50 >10.0g/kg.bw. Ames test and micronucleus test of bone marrow cell showed the sample of "Qixiang" aerosol was safe without mutagenesis. No toxicity was observed in sperm shape abnormality test in mice.2. "Qixiang" aerosol can promote obviously the phagocytic index of mononuclear macrophage and the Cytotoxic activity of splenic NK cell to raise nonspecific immunological function in immunosuppressive mice and have no obvious effect of nonspecific immunological function in normal mice.3. Compared with Cy group, "Qixiang" aerosol can significantly raise the half number hemolysis rate in serum mice(HC50) in Cy+"Qixiang" aerosol group. There is no obvious effect of humoral immune function in normal mice.4. Compared with Cy group, "Qixiang" aerosol can significantly promote T-lymphocyte function, the ability of IL-2 production by mice splenocytes, decrease the content of TNF-a induced by peritoneal macrophage in Cy+"Qixiang" aerosol group. There is obvious effect of cellular immune function in mice.
