多聚羟基烷酸异聚体的代谢调控及其下游加工过程研究

【摘要】 多聚羟基烷酸(PHA)是一类由生物合成的结构简单的高分子聚合物,它不仅具有石化塑料的特性,而且具有生物可降解性、生物相容性、降解产物无毒性的特性,是一种环境友好型的生物材料。这种新材料对于解决白色污染的环境问题以及医学组织工程框架材料有着重要的应用价值。 本论文研究了费氏中华根瘤菌S. fredii以葡萄糖为主要碳源,月桂酸和癸酸作为辅助碳源,通过代谢调控,分别获得了3-羟基丁酸和3-羟基辛酸共聚体P(HB-HO),3-羟基丁酸和3-羟基己酸共聚体P(HB-HH),并且分别研究了两种产品的高密度发酵调控实验的相关参数。其中聚合物P(HB-HO)填补了PHA系列生物材料的空白。实验结果表明:(1)P(HB-HO)中HO和P(HB-HH)中HH组分含量与月桂酸的加入时间和浓度有密切的关系,烷酸盐的最佳加入时间为胞内多聚物合成初期,即细胞生长期后期;尽管高浓度烷酸盐下可获得较高的HO、HH组分含量,但会明显抑制菌体的生长和产物的合成;通过控制烷酸盐浓度可以得到不同HO、HH组分含量的共聚体。(2)分别进行了P(HB-HO)和P(HB-HH)100L大罐发酵试验,在合成阶段,采用两阶段发酵方式和两步加盐法,不仅避免了高浓度烷酸盐对菌体的抑制作用,而且控制了冒顶现象。P(HB-HO)占干细胞最大为38.7%,单体比例为HO:HB=1:3.9,粘均分子量为1.51×105Da,P(HB-HH)总含量占干细胞30%,单体比例为HH:HB=1:1.6,粘均分子量1.37×10~5Da。 另外,分别进行了溶剂萃取法和水相分离法的两种方法提取PHA,通过正交数据分析设计实验,结果表明:使用氯仿-乙醇法提取2h-4h的效果最佳,收率可达到95.11%,纯度可达99.4%;而水相分离法中,硫酸法提纯产品纯度很低;碱法提取过程中,对产品的降解十分强烈;超声波法虽对细胞壁有一定的破碎作用,但是无法分离出与细胞残片混在一起的PHA颗粒。 接着我们对氯仿-乙醇萃取条件及不同萃取方式进行了研究,结果表明:(1)综合分析得出:萃取温度为40℃,氯仿与干细胞混合比例为5:1(v/g),处理2~4小时,萃取液浓缩到1/10体积,沉淀剂乙醇加量
【Abstract】 Polyhydroxyalkanoates(PHA) are polymers that are produced by many bacteria and accumulated as intracellular granules. It not only has the characteristic of petrochemical plastics but also has the biodegradability, biocompatility. These biologic polymers are new kinds of environment-friendly material that provide a novel way to resolve White Pollution and can be used as scaffold material in medical tissue engineering. Synthesis of the bioplastic poly (3-Hydroxybutyrate-co-3-Hydroxyhexanoate)[p(HBHH)],Poly(3-Hydroxybutyrate-co-Hydroxyoctanoate) [p(HBHO)] by a Sinorhizobium fredii strain using sodium decanoate and sodium dodecanoate as auxiliary carbon source,with Glucose as the primary carbon source. Due to the physical and biologic property of P(HB-HH) and P(HB-HO),they provide a wide prospect of application in medical tissue engineering.In this paper, principium process was established through the research on correlative parameter of metabolic regulation by two-stage fed-batch fermentation process in a 100-L fermentor system. The maximum P (HB-HH) production could be 10.34 g/L with a monomer ration of 61%(w/w) 3-HB and 30% (w/w) 3-HH.
