中药罗布麻叶及其有效部位的质量控制方法研究

【摘要】 药用罗布麻叶的原植物共有两属三种,即罗布麻属罗布麻,白麻属大叶白麻、白麻。本课题建立了罗布麻叶及其有效部位中总黄酮和金丝桃苷的含量测定方法,并对不同种属和产地的样本进行了定量、定性研究,并初步建立了药材与有效部位的质量标准。定量研究方面,本文首先对药材盐酸水解后苷元-槲皮素进行了HPLC含量测定。结果显示该法不能明显区别药典收载的品种-红麻与其混淆品。故进一步应用UV分光光度法建立罗布麻叶与有效部位中总黄酮含量的含测方法;应用HPLC 法建立罗布麻叶与有效部位中上述指标成分-金丝桃苷的含测方法,以上含测方法经方法学考察符合有关规定。应用所建立的方法对收集的样品含测结果显示,可以看出目前市场所售罗布麻叶的质量差异较大,制订罗布麻叶的质量控制方法意义重大。在定性研究方面,采用硅胶薄层、微乳薄层、HPLC 三种指纹图谱法对19 批样品进行鉴别研究。其中硅胶薄层指纹图谱法中以硅胶HF254 为吸附剂,以乙酸乙酯-丁酮-甲酸-水(6:3:1:1)为展开剂;微乳薄层指纹图谱法中以聚酰胺薄膜为吸附剂,以含水75%的微乳液为展开剂,这两种指纹图谱法均能将罗布麻属和白麻属准确区别开来,其中后者效果优于前者。HPLC 指纹图谱法以A:乙腈(含0.05%磷酸),B:水(含0.05%磷酸)为两元流动相梯度洗脱,以样16(对照药材)为对照模板,两属植物均出现了特有的色谱峰,罗布麻属出现18~38min 的特征区,白麻属出现特征性的8.9~20.0min 特征区域,可以容易地把药典收载品种-红麻鉴别出来。不同产地之间红麻相似度为0.88~1.00,罗布麻属与白麻属植物之间的相似度仅为0.5~0.6。本课题首次应用硅胶薄层、微乳薄层、HPLC 三种指纹图谱法对不同种属和产地的样品进行鉴别研究,首次应用HPLC 建立罗布麻叶及其有效部位中主要黄酮类成分-金丝桃苷含量测定方法,从药材与有效部位分别进行含量控制,更具有说服力。并在以上方法研究的基础上,建立罗布麻叶及其有效部位的质量标准,为罗布麻叶的进一步开发利用奠定了坚实基础。

【Abstract】 Luobuma (Apocynum venetum Line) was known as a kind of traditional Chinese medicine and used for treatment of hypertension, headache and blackout. Because of week quality control technique ,other plants from different sources,such as Poacynum hendersonii(Hook.f.)Woodson) and Poacynum pictum(Schrenk)Baill have been mistakenly used as Apocynum venetum Line for a long time.So, the quantificational and qualitative quality control methods of Luobuma leave and its active fraction had been estabalished and could be used as quality standard. The content of quercetin in hydrolysate of leaves had been determined by HPLC method firstly.But the result was found not to be exclusive enough.So,the UV method was established to determine the content of total flovonoids in Luobuma leave and its active fraction. Hyperin was selected as good representative consituent, which be colored by AlCl3-NaAc. Then, the HPLC method was established to determine the content of hyperin in Luobuma leave and its active fraction. All the methods of determination were accurate and presible. In this paper, total flovonoids and central active consituent-hyperin of samples from different species and areas were analysized by UV and HPLC methods respectively. Valuable conclusion had been drawn and great difference of content of total flovonoids and hyperin was found in samples of different species and areas. Exploratory researchs by chromatographic fingerprint of samples were done subsequently.Some useful harvests have been got by general TLC and microemulsion thin-layer chromatographic fingerprint. And the later shows advantage to discriminate Apocynum venetum L with its confused plants. The HPLC chromatographic fingerprint method also was established to reveal mass information from samples. Different unique peaks were found in samples from both species. And the particular section from 18 to 38min in the plants of Apocynum Linn. was found to be different from the unique section 8.9~20min in those of Poacynum Baill.. Based on those information mentioned above, Luobuma leave of different species could be identified easily. Analyzing by Computer Aided Similarity Evaluation System, the similarity of Luobuma leaves from the same aeras was 0.88~1.00, which from different species was 0.5~0.6. In a word, the content determination methods of total flovonoids and hyperin in Luobuma leave and its active fraction had been established firstly .Then three chromatographic fingerprint methods were applied on the samples from different species and areas. The experiment was a useful exploratory research.
