欧李快速繁殖技术体系研究

【摘要】 欧李(Prunus humilis(Bge).Sok)是蔷薇科樱桃属的小灌木植物,原产于我国,它是一种抗寒、耐旱、耐盐碱、耐贫瘠,适应性很强的野生果树。本研究以欧李的茎段为外植体进行组织培养体系的建立,通过随机区组设计,结合方差分析等,着重研究了欧李茎段培养的最佳培养基配方及培养程序,以及影响快速繁殖的关键性因素,初步建立了一套较为完整的欧李快速繁殖体系。主要的研究结果如下: 1.欧李快速繁殖的外植体以 5 月中旬至 6 月中旬取材好,其诱导的萌芽率高,污染率低。 2.启动培养中,培养基 L+ 1.0 mg/L BA + 0.5 mg/L NAA 的诱导效果最好,培养出的无菌苗长势好,生长快。 3.在增殖培养中,经过筛选得出,最佳培养基为 L+ 0.5 mg/L BA + 0.3 mg/L NAA,其增殖倍数高,植株生长势旺,适宜的 NAA 浓度为 0.1-0.5mg/L,BA 的浓度为0.5-1.0 mg/L,而 6-BA/NAA 的值在 1-10 之间对试管苗的增殖有利,继代周期以 35天为宜,合适的接种密度为 10 株/瓶。 4.生根培养中,培养基 L+ 0.5 mg/L LAA +3%蔗糖的生根效果最好,生根率高,生根苗质量好,生根苗以敞开瓶口加蒸馏水的炼苗方法比较适宜;在移栽时,可选蛭石或市售营养土为基质,其移栽成活率高。 5.在简化培养基的试验中,食用白砂糖、自来水、玻璃瓶分别可以取代分析蔗糖、蒸馏水、三角瓶,替代之后对试管苗生长和增殖等方面没有显著的影响。

【Abstract】 Prunus humilis, a species of Cerasus and originate in china,is cold-resistant, able to bear the drought、saline、alkalinly and barrenly, the wild fruit tree with very strong adaptability. The major object of this study is to exploit the technique systems of tissue culture and rapid propagation for Prunus humilis, through the randomised block design, combine analysis of variance, using the stems as the explant, and the optimal medium and culture programe are studied. The major results were as following: 1.Using the fresh stems of Prunus humilis as the explant for initial culture, the best season for selecting material was middle ten days from 5 to 6 kind soon. The germination ratio was high and less pollution. 2.The optimizing medium for stems initial culture was L+1.0 mg/LBA+0.5 mg/L NAA. The germination ratio was high with seedlings fast grow. 3. In subculture multiplication, the optimizing medium was L+0.5mg/LBA+0.3mg/L NAA.And high times of propagation, the tendency was prosperous that the plant grows. Suitable NAA density was 0.1-0.5mg/L, the density of BA was 0.5-1.0 mg/L, And the value of BA/NAA was useful to hyperplasia of the test tube shoot among 1-10,the period of subculture was suitable for 35 days, the suitable density of inoculation was 10 / bottle. 4.The optimizing medium of rooting was L+0.5 mg/LIAA+3%sugar, the ratio of rooting was high, with strong and quick-grow roots. With open wide bottleneck add to smelt shoot method suitable for relatively distilled water shoot that take root, While transplanting, we could choose vermiculite or nutrition that market sell soil host, the transplant survival rate was high. 5.In simplifying the test of the medium, white sugar, tap water and glass container, could replace and the A.R. sugar, distilled water and triangular bottle, and did not have remarkable influence on the test tube shoot after the substitution.
