阿魏侧耳子实体多糖分离纯化及其化学结构的初步研究

【摘要】 本文对阿魏侧耳子实体多糖的分离纯化及其级份PW2的化学结构进行了初步研究。阿魏侧耳子实体经过烘干、研磨粉碎后,再经热水提取醇沉得粗多糖PW。将PW用Sevag法脱蛋白后,用蒸馏水透析去离子再经DEAE-纤维素阴离子交换柱层析,分级分离得三种多糖级份PW1、PW2和PW3,颜色分别为淡黄色,白色,淡黄色。然后以多糖PW2为样品进行结构测定研究。 经紫外光谱鉴定多糖PW2不含有蛋白质和核酸;采用纸色谱和高压电泳以及柱色谱分析结果表明多糖PW2满足结构分析的要求。用高效液相色谱测定其分子量为3.18×10~4。多糖PW2经过完全酸水解后,用高效液相色谱分析结果表明该多糖是由葡萄糖和半乳糖两种单糖组成,摩尔比例为1.77:1。通过红外光谱,核磁共振测试分析,初步确定多糖PW2主要以α构型为主,含有少量β构型,单糖的连接方式主要以1—6连接为主链,含有1—4连接的支链,糖环构型为D-吡喃糖环。
【Abstract】 Isolation, Purification and Chemical Stucture ofPolysaccharide from Pleurotus ferulae LanziPostgraduate: li Jun; Director: Lu Zuo-zhou (Institute of Applied Mycology, Huazhong Agricultural University, Wuhan 430070)In this thesis, the isolation, purification and the structural characteristics of polysaccharide PW2 from the fruit body of Pleurotus ferulae Lanzi were studied. Mushroom powder was obtained by dry, grind after smashing from the fruit body, and was withdrawn by hot water and were precipitated by alcohol to get the protein-bond polysaccharide(PW). And then PW were taken off protein with the method of Sevag, used the distilled water dialysis to remove the ion. After DEAE-Cellulose ion change column chromatography, there were three polysaccharides PW1, PW2 and PW3, the color was thin and yellow respectively, white, thin and yellow. Finally the PW2 was regarded as the sample proceeding the construction measurement research.Passing UV spectrum, PW2 does not contain the protein and nucleic acid, and was proved to be pure through PC, gel filtration and electrophoresis. Its molecular weight were estimated to be 3.18 104 by HPLC. PW2 are completely hydrolyzed catalyzed by acid, to have the bond of indicant broken entirely and get the contents of sugar, measured by HPLC. PW2 are mainly made up of D-Glc and D-Gal by 1. 77:1 in molar. Analyzed by Infrared Ray(IR) and Nuclear Magnetic Resonance(NMR), The structure of PW2 is chiefly linked by a -glycoside bonds and a few glycoside bonds, while the backbone of PW2 is mainly linked by 1-6 glycoside bonds and a few 1-4 linkage.
