新疆维吾尔族长寿家族与非长寿家族DNA断裂水平及相关因素的研究

【摘要】 目的:研究新疆维吾尔族长寿家族与非长寿家族的DNA断裂水平及相关影响因素。方法:采用流行病学方法进行现场问卷调查,内容包括许多选择性条目,例如:生活方式、饮食习惯、吸烟饮酒史、遗传史、生育史、既往病史等等,同时对每一个调查对象进行体格检查,包括:身高、体重、血压、脉搏等,并且在征得本人同意后采集血液样本,通过彗星实验方法检测其血淋巴细胞DNA断裂水平。统计学分析采用t检验、秩和检验、相关分析、回归分析、协方差分析、X~2检验、Logistic回归等方法。结果:长寿家族组与非长寿家族组DNA断裂率均数分别为14.33%、21.93%,经统计学检验差别有统计学意义(P<0.01),而且DNA断裂不同分级之间也有统计学意义(P<0.01)。DNA断裂率与年龄成正相关,有随年龄增高而增加的趋势,不同年龄段(20-39岁、40-59岁及60-79岁)的DNA断裂率均数分别为11.18%、21.63%、23.17%,经统计学检验差别有统计学意义;把年龄作为一个协变量进行协方差分析发现两组的DNA断裂率校正均数仍然有统计学意义(P<0.01);男性、女性DNA断裂率均数分别为17.98%、17.19%,差别无统计学意义(P>0.05)。DNA断裂水平低、赤脚、饮用和田茶、常吃水果等可能是影响寿命的积极因素。结论:长寿家族与非长寿家族的DNA断裂水平的基础值存在明显差异,在排除年龄、性别、吸烟、饮酒、慢性病等因素的影响后,两组人群的DNA断裂率均数仍有统计学意义,依据衰老的DNA损伤修复学说,本次研究的两个人群组寿命的长短可能与遗传背景产生的DNA损伤不同有一定的关系;同时也与生活方式、饮食习惯有关。

【Abstract】 Objective: This paper is to study the background levels of DNA strand breaks and the correlative factors in Uygur nationality in Xinjiang. Method: By way of epidemiology, the questionnaire was conducted, including such aspects as life-style, diet habits, smoking and alcohol drinking history, heredity history, reproductive history and case history, etc. After getting 108 subjects’ permission, physical examination of each person was made, including height, weight, blood pressure and pulse. Meanwhile The blood samples were collected to evaluate DNA damage . in lymphocytes by using comet assay. Different statistical methods were employed, such as non-parametric test, t-test, correlation and regression analysis, covariance analysis, Crosstabes, Logistic regression and so on. Results: DNA damage frequencies between longevity families and non-longevity families are 14.33% and 21.93% , and they are statistically significant (P<0.01). In addition, the different grades of DNA damage are statistically significant, too (P<0. 01); DNA damage frequency is positively correlated with the age and will increase with the growing age. And its averages in different age groups (ages 20-39, 40-59 and 60-79) are 11. 18%, 21.63% and 23.17%, which are statistically significant(P<0. 01); the age was used as covariance to make covariance analysis, and it has been found that the remedy mean of DNA damage in the two groupswere still statistically significant (P<0.01). DNA damage frequencies of male and female were 17.98% and 17.19%, the difference was not statistically significant (P>0. 05). Logistic regression analysis found that low DNA damage Frequency as well as frequent bare feet, fruit taking, Hetian tea drinking constitute were the correlation factors of longevity. Conclusion: The non-longevity families have higher DNA damage Frequency than longevity ones and the difference is statistically significant. Excluding the factors of age, sex, smoking and alcohol drinking, etc, the mean of DNA damage Frequency in the two groups is still statistically significant . According to DNA damage and repair theory of aging, the life-span in the two groups may be related to the differences of DNA damage owing to different family backgrounds. Moreover, it has close relation with people’ s life-style and diet habits.
