电磁波控制热敏磁性脂质体药物释放前期研究

【摘要】 热敏磁性脂质体是由热敏脂质体、磁性材料和药物制备而成的一类脂质体。热敏磁性脂质体在肿瘤治疗方面同时具备多方面的优势,如选择性热疗、磁靶向给药系统等。由于它具有巨大的潜力和希望,国内外的学者正在努力使其可应用于医疗实践。为考察热敏磁性脂质体在整个电磁波段的电磁参数,研究Fe3O4超细粒子是必要而且重要的。在本文中,通过改变反应温度、pH值和反应物浓度以共沉淀方法制得了几组Fe3O4纳米粒样品。以X射线衍射谱分析了所制备样品组分为Fe3O4,以磁天平对Fe3O4纳米粒的比饱和磁化强度进行了表征。以PEG-4000和聚丙烯酸为分散剂制备Fe3O4磁流体的方法进行了探索。影响磁性药物微粒磁定位效果的因素包括磁性药物微粒的磁性能、血液动力学、磁场梯度以及肿瘤组织的不同特征。对磁性药物微粒的磁响应性、血液的特性以及血液动力学进行了详细分析,也考虑了磁性药物微粒浓度较高时的磁团聚力。对医疗中应用的各种电磁波频段以及它们在人体中的传播特性进行了整理归纳,从电磁学的角度对电磁波控制热敏磁性脂质体药物释放的机理进行了探讨,并建立了热敏磁性脂质体药物释放的数学模型。
【Abstract】 Thermosensitive magnetoliposomes are the kind of liposomes prepared with thermosensitive liposomes, magnetic materials and drugs. It is strongly suggested that thermosensitive magnetoliposomes can provide many advantages at the same time in treating cancer, that is, selective hyperthermia, magnetic targeted drug delivery systems and etc. With its great promise and potentiality in the future, scholars in home and abroad are managing to make the thermosensitive magnetoliposomes feasible in the medical practice.To investigate the electromagnetic parameters of thermosensitive magnetoliposomes in the whole electromagnetic waves frequencies band, studying super fine Fe3O4 nanoparticles is necessary and important. In this postgraduate thesis, several groups of Fe3O4 nanoparticles samples are acquired by the means of chemical co-precipitation changing the reacting temperature, pH and concentration. The component of Fe3O4 is verified with the X-ray diffraction spectra and the Specific Saturation Magnetization of each sample is characterized with the magnetic balance. Moreover, the method of preparing Fe3O4 magnetic fluid taking the PEG-4000 and polyacrylic acid (PAA) as dispersant is explored. The factors that influence the magnetic positioning include the magnetic properties of magnetic drug particles, hemodynamic forces, the magnetic filed gradients and variable tissue characteristics. The magnetic responsiveness of magnetic drug particles, the blood properties and the hemodynamic forces are analyzed in detail. The magnetic agglomeration force is considered when the magnetic drug particles are in high concentrations.The various electromagnetic frequencies applied in medical practice are listed and their spreading characteristics in human being are concluded. The principles of drug release of thermosensitive magnetoliposomes controlled by electromagnetic waves are discussed in the view of electromagnetics. The mathematical models of controlled drug release of thermosensitive magnetoliposomes are built.
