复合乌龙茶对雌性SD大鼠高脂血症血脂异常调节作用的实验研究

【摘要】 目的 建立高脂血症的动物模型,观察不同剂量复合乌龙茶对雌性SD大鼠高脂血症血脂代谢紊乱的预防效果,探讨其调节血脂的机制,为进一步将复合乌龙茶作为保健食品开发提供实验依据。方法1.通过药物筛选和专家咨询制定调脂复合乌龙茶组方;2.通过高脂饲料喂养建立高脂血症模型,测定血脂判断模型的建立;3.观察和评价不同剂量的复合乌龙茶对高脂血症大鼠血脂的调节功效,并探讨其机制。结果1.64天高脂饲料喂养后,高脂模型组大鼠血清总胆固醇(TC)、甘油三酯(TG)含量均较空白对照组显著升高 (p<0.05),而高密度脂蛋白(HDL)比空白对照组有显著下降(p<0.05),提示模型建立成功。2.中、高剂量复合乌龙茶组的TC、TG和LDL-C水平较高脂模型组显著降低(p<0.05),HDL-C显著升高(p<0.05)。乌龙茶组的TG较高脂模型组显著下降(p<0.05)。中剂量复合乌龙茶组与乌龙茶组相比较,前者可更有效地降低TC和LDL-C(p<0.05)。3.高、中剂量复合乌龙茶组动物肝脏胆固醇较高脂模型组显著下降(p<0.05);乌龙茶和各剂量复合乌龙茶组动物肝脏甘油三酯水平与高脂模型组相比无显著性差别(p>0.05)。4.高剂量复合乌龙茶组大鼠的肝脂酶(HL)活性较高脂模型组显著降低<WP=4>(p<0.05);高剂量复合乌龙茶组的脂蛋白脂酶(LPL)活性较高脂模型组显著下降(p<0.05)。结论通过向饲料中添加胆固醇,猪油,胆盐,可成功建立大鼠高脂血症模型。2.复合乌龙茶具有降低血清TC和TG,升高血清HDL-C功能。同等剂量的复合乌龙茶可比乌龙茶更有效地降低血清总胆固醇和低密度脂蛋白胆固醇。3.复合乌龙茶调节血脂代谢可能与增加大鼠粪便排泄,增加排泄粪便的脂肪含量,降低脂蛋白酯酶和肝脂酶的活性有关。

【Abstract】 ObjectiveThis study was conducted to explore the hypolipid role of multiplex oolong tea (MOT) on lipid metabolism in female SD rats with hyperlipidemia, and to gather the information for utilizing the multiplex oolong tea as functional food.Method1. The making of multiplex oolong tea is based on Chinese herbal medicine screening and counseling from specialist.2. 48 female SD rats of 2 months old were divided randomly into 6 groups by body weights: (I) control group, (II) hyerlipidemia model group, (III) MOT of high dosage, (IV) MOT of middle dosage, (V) MOT of high dosage and (VI) oolong tea group. Rats in group II-VI were fed with fat-enriched diet for 64 days. Lipid profile was checked to make sure that the diet-induced hyperlipidemia was formulated. 3. Body weight, serum total cholesterol (TC), triglyceride (TG), LDL-C, HDL-C, liver cholesterol (LC), liver triglyceride (LG), hepatic lipase and lipoprotein lipase activity, feces excretion and fecal fatty content were examined to evaluate and probe the modulatory role of multiplex oolong tea.Result1. TC, TG, LDL-C level were elevated in hyperlipidemia model group compared with those in control group (p<0.05). However, the HDL-C concentration is significantly decreased in model group compared with that in control group (p<0.05 ). 2. Compared with hyerlipidemia model, serum TC and TG of IV and V groups were <WP=6>significantly reduced (p<0.05). TG in group VI is lower than that of model group (p<0.05). TC and LDL-C level in MOT of middle dosage is significantly reduced than those treated with oolong tea (p<0.05). 3. LC content of group IV and V were decreased compared with that model group (p<0.05), however, the LG level in group III-VI didn’t differ with that of model group (p>0.05).4. HL and LPL activity of group V were reduced compared with model group (p<0.05). Conclusion 1. SD rats fet with diet rich in cholesterol, lard and bile acid salt for 64 days will formulate hyperlipidemia.2. MOT can lowing TC and TG, elevating HDL-C. The effect of lowing TC and LDL-C of MOT is better than that of oolong tea.3. MOT may modulate the lipid level through increasing feces excretion and elevating fecal content as well as depressing HL and LPL activity.
