新扬州鸡DNA指纹J带及两个SCAR标记与生产性能的综合效应研究

【摘要】 本研究以新扬州鸡为试验素材,利用DNA指纹和SCAR技术对样品进行研究。DNA指纹技术中,以EAV(禽内源性反转录病毒片段)为探针,以EcoRI为限制性内切酶,进行DNA指纹检测,探讨DNA指纹图谱中长度为3.48kb的条带J对鸡群8周龄体重、10周龄体重、26周龄体重的影响。SCAR标记技术中,通过优化OPAY02随机引物的PCR反应,得到清晰的图谱,回收1660bp和2326bp的条带,进行克隆,测序,设计两对特异性引物,分别对新扬州鸡群基因组DNA进行扩增,然后结合两种SCAR标记与J带,利用广义线性模型,进行综合性分析。结果表明:DNA指纹J带同样在新扬州鸡群中存在,且对新扬州鸡8周龄体重、10周龄体重、26周龄体重均有显著影响;SCAR标记分析中,发现S2标记对三个周龄体重影响均达到显著增效效应,而S1标记也表现出一定的减效效应,但并不稳定,仅对26周龄体重存在显著效应,但结合其它标记,发现S1标记效应明显;在标记组合效应中,得到S2+S1-、J-S2+、J-S1-及J-S2+S1-标记组合型对新扬州鸡群三个周龄体重影响均为显著,可作为遗传标记,并得到最佳组合J-S2+S1-;标记互作效应分析中,J带、S1标记、S2标记与性别两两可组合间均无显著互作效应。综合分析结果,可选择J带、s:标记、52+s,一(S。)、丁52+、J’s:.及J’52+5,一作为新扬州鸡的分子遗传标记,对鸡群进行早期选择。

【Abstract】 DNA fingerprinting and SCAR procedures were carried with New Yangzhou chickens used as experimental animal. General Linear model (GLM) was used for the analyses of the relations between the markers and productive performance. The random primer OPAY02 was used in order to recover targeted fragments (1660bp and 2326bp, respectively). Two pair of primers were designed through cloning and sequencing procedures. The designed primers were then used for SCAR-PCR with New Yangzhou chickens DNA samples serving as the template. The results are as follows:The effects of band J on productive performance, including 8,10 and 26-week weights was analysed by DNA fingerprinting with Endogenous avian virus (EAV) used as the probe. The results demonstrated that band J exists in genome of New Yangzhou chickens and band J had significant influence on 8,10 and 26-week weights, respectively. The productive performance of the chickens without band J were higher than those with bandJ.The analyses of SCAR markers showed that S2-marker has significant effect on the three productive traits and Si-marker only has significant effect on 26-week weight Butas Si-marker is related to the other markers, it shows significant effects.In the. analyses of compositive effects, have significanteffects on the three productive traits and the optimum composite depicted as Jwas obtained.In the analyses of interactive effects, Band J, S1 and S2-markers and sex were foundwith no interaction between two factors.This investigation revealed that Band J, obtained can be used in marker assistant selection (MAS), to minimise cost, speedselection progress and stock-breeding.
