豹皮樟总黄酮对非酒精性脂肪性肝炎的防治作用及部分机制研究

【摘要】 非酒精性脂肪性肝炎是以无过量饮酒史以及肝细胞脂肪变性、气球样变、弥漫性肝小叶炎症和(或)肝中央静脉、肝窦周围胶原沉积为特征的慢性肝脏疾病。目前临床上主要使用调血脂和改善胰岛素抵抗的药物进行治疗,但疗效和安全性尚需进一步评价,且暂无特效治疗药物。 豹皮樟,具有清凉止渴、解毒消肿、明目健胃等功效,但迄今未见该植物的药理作用报道。为此,本项目组经过多次实验,确定了该植物治疗非酒精性脂肪性肝炎的有效部位为黄酮类。 本文采用脂肪乳剂灌胃诱导实验模型,并研究了TFLC对非酒精性脂肪性肝炎的防治作用及部分作用机制。主要内容如下: 1.建立非酒精性脂肪性肝炎模型 通过脂肪乳剂灌胃,大鼠血和肝脏TG升高,病理显示肝细胞脂肪变性,并出现明显炎症病灶。提示,模型诱导成功。 2.TFLC对非酒精性脂肪性肝炎的防治作用 造模同时给予TFLC,TFLC能降低大鼠TG水平,改善脂肪变,减轻炎症。其机制可能是:抑制TG在肠道的吸收;减轻脂质过氧化。 3.TFLC对非酒精性脂肪性肝炎的治疗作用及部分机制 造模成功后通过整体和体外给药,TFLC能降低大鼠TG水平,改善脂肪变,减轻炎症。其机制可能是:降低FFA和TNF-α对肝脏毒性;减轻脂质过氧化。

【Abstract】 To explore the effects of TFLC on nonalcoholic steatohepatitis(NASH), NASH model was adopted by fat emulsion.The mechanisms of action of TFLC on NASH by using the methods in vivo and in vitro at body,cellular levels were studied.The main contents are divided into some sections as follows: 1.To establish the NASH animal modelThe serum and hepatic TG increased markedly by fat emulsion in rats,and the histopathology research showed hepatocellular macrovesicular steatosis .The results indicated the experimental animal NASH in rats were successfully established . 2.To research preventive effects of TFLC on NASH.The rats were fed with fat emulsion and TFLC at the same time.TFLC markedly reduced the elevated level of serum and liver tissue TGLiver histopathological examination showed TFLC could ameliorate hepatocellular macrovesicular steatosis.The protective and therapeutic action of TFLC on NASH would be associated with inhibitory effect of fat absorption in the digestive tract and lipid peroxidition.3.To research therapeutic effects of TFLC on NASH, and its partly mechanisms of actionThe rats were first fed with fat emulsion for 3 weeks .Added to drugs by in vivo and in ivtro,TPLC markedly reduced the elevated TG level of serum ,liver tissue and HC.Liver histopathological examination showed TFLC could ameliorate hepatocellular macrovesicular steatosis.The therapeutic action of TFLC on NASH would be associated with reduction of FFA,TNF- a and lipid peroxidition.
