陈皮中抗肿瘤活性成分的研究

【摘要】 本论文目的在于研究陈皮药材中的抗肿瘤的化学成分及主要活性成分的含量测定。采用硅胶层析技术和大孔树脂吸附技术分离陈皮提取物,运用1HNMR,13CNMR. FAB-MS等光谱技术和化学方法鉴定得到的单体;运用高效液相技术进行含量测定。 在有效部位中分离得到了10个化合物,鉴定了其中7个化合物。其中黄酮5个,分别为3,5,6,7,8,3′,4′-七甲氧基黄酮、川陈皮素、5-羟基-3′,4′,6,7,8-五甲氧基黄酮、3,5,6,7,3′,4′-六甲氧基黄酮和橙皮甙。另外两个是醋酸甘油酯和β谷甾醇。建立了陈皮中3,5,6,7,8,3′,4′-七甲氧基黄酮和川陈皮素的定量分析方法,实验结果表明3,5,6,7,8,3′,4′-七甲氧基黄酮在0.025-0.4mg/ml范围内呈良好线性关系,相关系数r=1,平均加样回收率98.74%(n=5),RSD为0.93%。川陈皮素在0.025-0.4mg/ml范围内呈良好线性关系,相关系数r=0.9998,平均加样回收率97.5%(n=5),RSD为2.6%(n=5)。测定三个产地的3,5,6,7,8,3′,4′-七甲氧基黄酮和川陈皮素的含量。 药效学实验表明陈皮中特有的多种低极性部分的聚甲氧基黄酮类化合物具备一定的抗肿瘤活性,并且无毒副作用,是很有前途的抗肿瘤天然药物。含量测定结果表明陈皮中有抗肿瘤活性的3,5,6,7,8,3′,4′-七甲氧基黄酮和川陈皮素含量随产地不同变化很明显,作为道地药材的广陈皮中3,5,6,7,8,3′,4′-七甲氧基黄酮的含量较其它两地高。这些结果为陈皮抗肿瘤的研究和陈皮药材的质量标准的制定提供了参考。
【Abstract】 The effective fraction of Citrus reticulata Bianco are found to have the effect of anti-tumor. The purpose is to study the anti-tumor active ingredient from Citrus reticulata Bianco . The compounds are isolated with silica gel and compounds are identified with UV, MS, 1HNMR,13CNMR spectra. The content of 3,5,6,7,8,3’,4’- heptamethphoxyflavone and nobiletin are determined by HPLC.the effects of anti-tumor active ingredient from Citrus reticulata Bianco are extracted and isolated. 10 compounds are isolated with silica gel and compounds are identified with UV, MS, 1HNMR,13CNMR spectra. They are nobiletin , 3,5,6,7,8,3’,4’- heptamethphoxyflavone , hesperidin, -sitosterol, 5-hydroxy3’,4’,6,7,8-hexadicflavone,3,5,6,7, 3’,4’- hexadicflavone, glyceric acetate.HPLC figs of the effects of anti-tumor dementia 3,5,6,7,8,3’,4’- heptamethphoxyflavone and nobiletin from Citrus reticulata Bianco is established primarily. The result show that the linear range of 3,5,6,7,8,3’,4’- heptamethphoxyflavone is 0.025-0.4mg/ml, the correlation coefficients is 1 and the linear range of nobiletin is 0.025-0.4mg/ml, the correlation coefficients is 0.9998. The average recoveries of 3,5,6,7,8,3’,4’- heptamethphoxyflavone and nobiletin respectively are 98.74% (n=5,RSD=0:93%), 97.5%(n=5, RSD=2.6%).In a conclusion, Citrus reticulate Bianco has a lot of special flavone. They have certain anti-tumor activity. I have determined the content of 3,5,6,7,8,3’,4’- heptamethphoxyflavone and nobiletin from three locations. The result show that the content of 3,5,6,7,8,3 ’,4’-heptamethphoxyflavone and nobiletin from Guangzhou are much higher than the other two.
