苏太猪肉质性状与15号染色体微卫星标记相关性研究

【摘要】 对100头苏太肥育猪进行屠宰测定,测定指标包括:胴体重、背膘厚、嫩度、大理石纹、系水力、pH值和肌内脂含量,其统计分析结果表明:苏太猪背膘较薄,2.58±0.57cm,大理石纹评分优良3.24±0.57,肌肉pH值为6.09±0.19。值得注意的是:苏太猪的嫩度和肌内脂含量的变异度很大。采用SPSS对表型性状进行相关分析表明:胴体重与背膘厚之间极显著相关(r:0.569),胴体重与大理石纹、pH值、嫩度之间间显著相关(r分别为:0.223,-0.253,和-0.233);背膘厚与嫩度、肌内脂含量之间极显著相关(r分别为:-0.342,0.319),背膘厚与大理石纹间显著相关(r:0.255)。大理石纹与嫩度显著负相关(r=-0.260)、与肌内脂含量极显著正相关(r=0.510)。嫩度与pH值显著正相关(r=0.213),而与肌内脂含量显著负相关(r=-0.250)。 选取家猪15号染色体上的6个微卫星标记在苏太猪猪群内进行多态性检测。结果表明,6个微卫星位点均为多态性座位,各座位扩增片段大小从92bp到302bp不等。6个基因座等位基因数目为3-7个,各个基因座等位基因频率在0.01-0.57之间,杂合度为0.52-0.79之间,多态信息含量为0.42-0.76。6个微卫星的平均杂合度为0.71,平均多态信息含量为0.66。由此可见,所选择的微卫星座均为中高度多态性基因座。 利用SAS的GLM过程分析微卫星标记与肉质性状间的相关性,结果表明微卫星微卫星GAD(11323)和S0149(10819)均与pH值显著相关,S0088(10208)与嫩度显著相关、与系水力极显著相关,S0284(11242)与pH值和系水力均极显著相关。利用SPSS单向方差分析中的LSD法对相关显著的微卫星和肉质性进行多重比较,结果表明:与肉质性状相关显著的4个微卫星其多个基因型之间有显著差异,这4个微卫星可以作为苏太猪进一步选育的参考标记。

【Abstract】 100 SuTai fattening pigs were slaughtered and their carcass weight, BFT (backfat thickness), tenderness, marbling, WHC (water-holding capacity), pH value and IMF (intramuscular fat content ) were measured. The result of descriptive statistics shows that the BFT of Sutai pig is very thin (2.58 0.57cm) and the marbling score is favorable(3.24 0.57) and the pH value (6.09 0.1 9) of muscle coincide with that reported. But it is notable that the tenderness and the IMF, important to pork quality, vary greatly. According to the analysis by SPSS, the correlation between different traits is significant or extremely significant between carcass weight and BFT, marbling, pH value, tenderness (r: 0.569**, 0.223*, -0.253*, -0.233*); BFT and tenderness, IMF, marbling (r:-0.342**, 0.319**, 0.255*); marbling and tenderness, IMF(r: -0.26* , -0.51*); tenderness and pH value, IMF(r:-0.213*, -0.25*).Six microsatellites on chromosome 15 were selected to investigate polymorphism in Sutai pig population by PCR and PAGE electrophoresis. The result shows that 6 microsatellites are polymorphic and the fragments of PCR vary from 92bp to 302bp. The allele number varies from 3 to 7, and the frequency of allele from 0.01 to 0.57, heterozygosity from 0.52 to 0.79, polymorphism information content (PIC) from 0.42 to 0.76. Mean heterozygosity of the six microsatellites is 0.71 and mean PIC is 0.66.Thus it can be seen that the microsatellites are medial or high polymorphic loci.The correlation between the microsatellite markers and pork quality traits were analysed by SAS-GLM procedure. The results showed that there were significant correlation between microsatellite GAD (11323 ), 80149(10819) and pH value, bewteen S0088 and tenderness , WHC (extremely significant), extremely significant between S0284 and pH value, WHC. The analysis by LSD of one-way ANOVA in SPSS on those microsatellites significant correlative with pork quality traits shows that there is significant difference between the genotypes of 4 microsatellites. Thus these microsatellitesmay be referenced as the markers for further breeding of Sutai pig.
