参术多糖GMPS的初步研究

【摘要】 红参、白术是传统的补益中药。本课题是以中医药理论为指导,在药效学研究的基础上,寻找参术总多糖中治疗胃癌癌前病变的有效部位GMPS,并对GMPS的提取纯化方法、理化性质、组分分析等进行了初步药学研究。研究内容如下: 水煎提取参术总多糖,并运用分段醇沉法将其分为四个部位,分别用于药效学研究,筛选出治疗胃癌癌前病变的有效部位GMPS。 对GMPS的提取条件进行了研究:药材经醇提除杂后,以GMPS收量为指标,采用L9(3)4正交设计,对GMPS水提取工艺进行考察,确定其最佳水提工艺为加6倍量水煎煮3次,每次1.5小时;并结合分段醇沉法,收集30%~60%醇沉部位即GMPS。同法收集红参、白术中与GMPS相同的醇沉部位即GPS、MPS。 对GMPS的性质进行了初步研究:本品为灰白色至灰色粉末,无味。对GMPS的水解条件进行了考察,应用TLC、PC对GMPS所含的组分进行了初步探索,并与GPS、MPS进行了比较。结果表明GMPS含有GalA、Gal、Xyl、Rha,不能肯定是否含有Ara、Glu、Man。无法确定GMPS中各组分的归属。 对除蛋白方法进行了比较,选定TCA法。GMPS除蛋白后,经DE-52纤维素柱层析分离得四个亚级分。同法分离GPS、MPS。对GMPS中各亚级分的红外光谱作了分析,并与GPS、MPS中相同洗脱部位的亚级分进行了比较。结果表明GMPS的组成成分与GPS、MPS有差异,提示混煎多糖不同于单煎多糖的混合物。

【Abstract】 Radix ginseng rubra and Rhizoma atractylodis macroceophaiae are topic traditional Chinese medicines. This research is on the base of traditional Chinese medicine-theory and pharmacodynmics study, using modern analysis technology find GMPS having effect on pathology of pregastric cancer from Radix ginseng rubra and Rhizoma atractylodis macroceophaiae and initially research on GMPS extracting, purifying method, physical-chemical character and component analysis of the function parts. The content of this research:Use water boilingRadix ginseng rubra and Rhizoma atractylodis macroceophaiae polysaccharide and get 4 parts precipitated by ethanol. Study the pharmacodynamics of these 4 parts respectively to screen effective parts GMPS which is effective to pregastric cancer.Study on GMPS extracting condition, after the medicinal materials extracted by ethanol. Use the quality of GMPS as the target to investigate water-extracting condition of GMPS with taking orthogonal L9 (3)4 design. Determinate the best water-extracting procedure: add 6 time water to boil 3 times, 1.5 hours each and combine with ethanol precipitated method. The 30%-60% ethanol precipitated parts is GMPS. The same method collects GPS, MPS, the ethanol precipitated parts same as GMPS from Radix ginseng rubraRhizoma atractylodis macroceophaiae.Tentative study on GMPS character: grayish-white to gray powder, odourless. Investigate hydrolysis condition. Use TLC, PC to initially probe into to components GMPS contain. Compare it with GPS and MPS. The result shows that GMPS contains Gal, Xyl, Rha, Gal A; can’ t be sure whether have Ara, Glu, Man; can’ t know the source of the parts in GMPS.Choose method of TCA by comparing with deproteinization method. After deproteinization GMPS through DE-52 cellulose column chromatography we can get three secondary components. The same way separates GPS, MPS. Analysis the secondary parts in GMPS by infrared spectrophotometer and compared with secondary parts the same wash parts in GPS, MPS. Result shows that the component in GMPS is different to GPS, MPS which include that mixed-boiling polysaccharide differ to separate-boiling polysaccharide mixed together.
