蝉蜕提取物诱导活动期SLE患者淋巴细胞凋亡的实验研究

【摘要】 目的:探讨蝉蜕提取物对活动期SLE患者淋巴细胞的作用。 方法:应用体外细胞培养技术,以500ug/ml~4000ug/ml蝉蜕提取物作用于体外培养的活动期SLE患者淋巴细胞,同时设空白组,CTX200ug/ml阳性对照组,共同温育12h、24h、48h、72h后,用MTT比色法计算细胞存活率,运用流式细胞仪分析细胞凋亡率,并检测CD69表达率。 结果:(1)500ug/ml~4000ug/ml蝉蜕提取物可诱导活动期SLE患者淋巴细胞凋亡,随着蝉蜕提取物浓度增大和作用时间的延长,凋亡细胞增多,呈现剂量依赖效应。(2)检测,T淋巴细胞CD69表达率,在24h时达39.1%±1.6%。 结论:蝉蜕提取物有促进活动期SLE患者淋巴细胞活化后凋亡的作用。

【Abstract】 Objective: To study of apoptosis induced in lymphocyte from patients with active Systemic Lupus Erythematosus (SLE) by Cryptotympana Pustulata extract(CPE). Methods: By techniques of cell culture in vitro, lymphocyte from patients with active SLE were treated by CPE in 500ug/ml~4000ug/ml concentrations for 12h, 24h, 48h, 72h. Meanwhile two control groups were set up including normal group, CTX 200ug/ml. Then computed ratio of the cell survival by MTT. Observed and analyzed the ratio of the cell apoptosis, CD69 of expression by Flow-cytometry. Results: (l)500ug/ml~4000ug/ml CPE could induce lymphocyte from patients with active SLE apoptosis. The concentration of CPE showed a dose-and-time dependent effect.(2)The ratio of CD69 expression is 39.1%+1.6% when treated with 4000ug/ml CPE after 24h. Conclusion: It is found that CPE could induce lymphocyte from patients with active SLE activation to apoptosis.
