肿瘤细胞染色体着丝粒点(Cd)变异的研究

【摘要】 目的:染色体着丝粒点(centromeric dots, Cd)指位于染色体着丝粒区、2条姊妹染色单体的外侧表层部位,并能被特殊方法着色的一对球形结构。研究染色体着丝粒点(Cd)的染色方法,分析及评价两种染色方法对染色体着丝粒点(Cd)的染色效果。 方法:分别应用Giemsa法和染色体着丝粒点—核仁组织区(Cd-NOR)同步银染法对人类外周血淋巴细胞中期染色体标本进行染色。 结果:(1) Giemsa法操作简便,但稳定性不好,Cd结构的清晰度差。(2)Cd-NOR同步银染技术方法简单,稳定性好,Cd带纹结构清晰。结论:Cd-NOR同步银染技术操作简便易行、方法稳定、所需试剂价格低廉,在一般光学显微镜下就可观察,一般实验室条件均能开展,制作的标本片易长期保存,有较大的实际应用价值,适宜基层推广应用。

【Abstract】 OBJECTIVE: Chromosome centromeric dots are a pair of spherical structure that can be painted by special staining methods, which lies in the centromere of two chromatid’s outer space. To study on staining methods of chromosome centromeric dots, to analyse and evaluate effect of two staining methods.METHODS: Giemsa method and Simultaneous silver staining technique of both NOR and Cd are used to stain metaphase chromosome specimen of human outer circular blood respectively.RESULTS: (1) Giemsa method is operated conveniently, but its stability is not good, structural definition of Cd is bad. (2) Simultaneous silver staining technique of both NOR and Cd is operated easily, its stability is good, structural definition of Cd band is good.CONCLUSION: Simultaneous silver staining technique of both NOR and Cd is operated easily, its stability is good, structural definition of <WP=8>Cd band is good. Its required reagent are cheap, it can be observed with a common optical microscope, thus it can be done in general laboratory Condition. Its specimen can be preserved long, Simultaneous silver staining technique of both NOR and Cd has a biggish applicable value practically.
