荧光试剂N-9-吖啶基-α-氨基酸及其甲酯的合成、表征和应用

【摘要】 本论文的主要工作分为两个部分:第一部分,用不同的方法优化合成了新型荧光试剂5a~5i:N-9-吖啶基-L-氨基乙酸、N-9-吖啶基-L-氨基丙酸、N-9-吖啶基-L-颉氨酸、N-9-吖啶基-L-苯丙氨酸、N-9-吖啶基-L-甲硫氨酸、N-9-吖啶基-L-天冬氨酸、N-9-吖啶基-L-半胱氨酸、N-9-吖啶基-L-苏氨酸和N-9-吖啶基-D-颉氨酸。合成了6a~6i:N-9-吖啶基-L-氨基乙酸甲酯、N-9-吖啶基-L-氨基丙酸甲酯、N-9-吖啶基-L-颉氨酸甲酯、N-9-吖啶基-L-苯丙氨酸甲酯、N-9-吖啶基-L-甲硫氨酸甲酯、N-9-吖啶基-L-天冬氨酸甲酯、N-9-吖啶基-L-半胱氨酸甲酯、N-9-吖啶基-L-苏氨酸甲酯和N-9-吖啶基-D-颉氨酸甲酯。产物均通过红外、核磁共振、旋光度和元素分析等进行了结构表征。另外,采用微波辐射法,简便、快捷地合成了7种N-苯基-α-氨基酸9a~9g,优化了经典的Bernthsen方法。 第二部分,对目标化合物N-9-吖啶基-α-氨基酸5a~5i的荧光和紫外光谱性质以及N-9-吖啶基-α-氨基酸甲酯5a~5i的水解性质进行了初步探讨和研究。 在此基础上选择性地将N-9-吖啶基氨基乙酸用于对痕量铜离子的荧光分析研究。结果表明,N-9-吖啶基氨基乙酸是一种高效率、高选择性、重现性好的测定Cu2+的荧光试剂。
【Abstract】 This paper is composed of the following two parts: In the first part, new fluorescent reagents 5a~5i were synthesized in different optimized methods. The reagents are: N-9-acridinyl glycine, N-9-acridinyl-L-alanine, N-9-acridinyl-L-valine, N-9-acridinyl-L-phenyl -alanine, N-9-acridinyl -L-methionine, N-9-acridinyl-L-aspartic acid, N-9-acridinyl-L-threonine, N-9-acridinyl-L-cysteine and N-9-acridinyl- -L-cysteine. 6a~6i were also synthesized: N-9-acridinyl glycine methyl ester, N-9-acridinyl-L-alanine methyl ester, N-9-acridinyl-L-valine methyl ester, N-9-acridinyl-L-phenylalanine methyl ester, N-9-acridinyl- -L-methionine methyl ester, N-9-acridinyl-L-aspartic acid methyl ester, N-9-acridinyl-L-threonine methyl ester, N-9-acridinyl- L-cysteine methyl ester and N-9-acridinyl-D-valine methyl ester. All the compounds were characterized by IR, 1H-NMR and element analysis. Besides, some N-phenyl-α-amino acids were synthesized by microwave radiation. This method was simple and convenient.In the second part, fluorescence and UV spectra of the aimed compounds 5a~5i were studied. The hydrolyzed property of 6a~6i was studied, too. On the base of this, N-9-acridinyl glycine was applied in the fluorescent determination of Cu(Ⅱ) and it shows that the method is simple, rapid and the results are reproducible.
