猪瘟免疫程序的研究

【摘要】 目的:为了探讨一种比较适合我省的猪瘟合理免疫程序,进行本实验。 方法:用不同免疫时间、免疫剂量对仔猪及母猪进行免疫,然后采集血清,检测猪瘟抗体水平。 结果: 1.仔猪母源抗体的消长:母猪配种前5头份免疫,至分娩前两周抗体保持在8以上,其所生仔猪的母源抗体在出生后逐渐上升,至20日龄达到最高5,随后逐渐下降,至30日龄抗体下降至3.9,至50日龄基本消失。 2.超前免疫场将二免时间从60日龄提前至45日龄,二免剂量从1头份提高至4头份,在80日龄,猪瘟抗体水平由原来的2提高到4.2(P<0.01)。 3.在农业部建议的常规免疫程序中,20日龄首免剂量由2头份提高到4头份,在40日龄时,猪瘟抗体水平由2.3提高到5.8(P<0.01)。60日龄二免剂量由4头份提高到6头份,在80日龄时,猪瘟抗体水平由1.7提高到5.0(P<0.01)。 4.经产母猪配种前用4头份、5头份、6头份进行免疫,至分娩前两周抗体水平分别为8.6、7.75、7.13,4头份组与6头份组差异显著(P<0.05)。 5.总计检测了福建省7个规模化猪场932份血清,只有516份血清猪瘟抗体达到阳性水平(IHA≥1∶16),阳性率55%。其中母猪血清检测了67份,100%达到阳性水平;哺乳仔猪183份,阳性率75%;断乳仔猪340份,阳性率53%;育肥猪阶段348份,阳性率只有39%。 结论: 1.猪瘟的超前免疫剂量2头份,根据抗体水平下降情况在45~50日龄进行4头份剂量二免较为合适。 2.常规免疫20日龄一免剂量4头份,60日龄二免剂量6头份较为合适。 3.经产母猪配种前进行免疫,4头份剂量即可取得良好免疫效果,不必使用过大的剂量。 4.调查福建省部分规模化猪场猪瘟抗体水平,发现猪群中猪瘟抗体水平普遍偏低,抗体阳性率只有55%,而且各场猪群的抗体水平不一。因此,必须重视抗体水平的监测,然后根据各场的具体情况制定个性化的免疫程序。注:1.本文的猪瘟抗体水平即猪瘟抗体滴度,用以2为底的对数1092表示,如猪瘟抗体滴度为l:64,用1092胡表示,简写为6。2.文中提到的抗体阳性水平指猪瘟抗体IHA滴度)1;16,抗体阳性率指抗体在阳性水平以上的占总检测数的比率。3.本实验中各规模化猪场所用猪瘟疫苗除了有两个场(③场和④场)使用哈尔滨兽医研究所生产的牛体反应苗外,其余场均使用福建省生物药品厂生产的组织苗。
【Abstract】 Purpose: In order to discuss a kind of reasonable immunity procedure for Hog Cholera that suited for our province, we did this experiment. Means: Vaccinate piglets and sow by different immunity time and dosage, then collect sera, detect Hog Cholera antibody level. Results:1. Eat and flow of materal antibody in piglets: Antibody level of sow that were vaccinated by 5 doses before hybridization keep higher than 1: 256. Materal antibody raised gradually after piglets were born, run up to maximum at 20 days old, then fell off subsequently. Antibody had protective action at 30 days old, disappeared basically at 50 days old.2. Advanced led immune piglets’ second vaccination time and increase dosage to 4 doses, antibody level mount up to 4.2 from 2(P<0.01) at 80 days old.3. In the routine immunity procedure that suggested by Agriculture Ministry, dosage of first vaccination at 20 days old increase to 4 doses from 2 doses, Hog Cholera antibody level mount up to 5.8 from 2.3 (P <0.01) at 40 days old. Dosage of second vaccination at 60 days old increase to 6 doses from 4 doses, Hog Cholera antibody mount up to 5.0 from 1.7 (P<0.01) at 80days old.4. Multiparity sow were vaccinated by 4, 5, 6 doses before hybridization, Hog Cholera antibody level were 8.6, 7.75, 7.13 respectively at two weeks before childbearing. Difference between 4 group and 6 group is very prominence (P<0.01 ) .5. Detected 932 copies of sera specimen from 7 scale hog pen in Fujian Province, thereinto Hog Cholera antibody of 492 copies run up to positive level (IHA > 1 : 16) , positive percentage was 55%. Thereinto detected 67 copies of sow sera, 100% run up to positive level; 183 copies of suckling pigs, positive percentage was 75%; 340 copies of ablactation pigs, positive percentage was 53%; 348 copies of finishing pigs, positive percentage was only 39%.Conclusion:1. Led immunizing by 2 doses, second vaccination at 45-50 days oldby 4 doses is appropriate according to the antibody falling instance.2. First vaccination of routine immunity at 20 days old by 4 doses, second vaccination by 6 doses at 60 days old is appropriate.3. Multiparity sow vaccinated before hybridization by 4 doses can get well immunity effect, do not need to use too much dosage.4. Detected Hog Cholera antibody level from part of scale hog pen, discovered that antibody level was on the low side and positive percentage was only 55%. We must set a high value on antibody inspection and map out individuation immunity procedure.
