干细胞和胶原蛋白结构性质的AFM研究

【摘要】 运用原子力显微镜(AFM)和扫描电子显微镜(SEM)这两种技术对BALB/c小鼠胚胎干细胞的细胞膜表面进行了观测,同时运用透射电子显微镜(TEM),通过制作超薄切片对BALB/c小鼠胚胎干细胞的内部结构进行观测,得到BALB/c小鼠胚胎干细胞的形态学特征。同时将三种技术进行比较,结果显示各有所长,应将这种新的技术(AFM)与传统技术(SEM与TEM)综合利用,互相补充。 利用AFM观测正常的CD34+造血干细胞,得到了单个造血干细胞以及膜表面超微结构的AFM图像,同时应用AFM测定正常人终末分化细胞红细胞的细胞膜表面超微结构,通过五个参数(Rp-v、Median height、Mean height、Rrms及Ra)定量比较造血干细胞与红细胞膜的特征,通过形态学的比较为进一步认识多能造血干细胞演变为终末血细胞的过程提供实验依据。 利用AFM研究不同种属Ⅰ型胶原蛋白聚集行为的差别,结果发现相同实验条件下同型胶原蛋白溶液中,胶原蛋白分子的聚合方式有所区别。来源不同,胶原蛋白分子聚集形成的胶原纤维在微观结构上有较大的差异,本实验结果对于胶原制品的开发及改善胶原制品的性能具有一定的参考意义。 以胶原蛋白为研究对象,对NSOM的成像条件进行摸索,通过对比实验说明探针的质量及定位系统对图像质量的影响。得到了自组装后胶原蛋白的不同扫描范围的透射模式近场光学像以及反射模式近场光学像。将NSOM技术与AFM技术进行了对比。

【Abstract】 BALB/c murine embryonic stem(ES) cell membrane was observed by atomic force microscope(AFM) and scanning electron microscope(SEM) and the inner structure of ES cells was studied by transmission electron microscope(TEM)in order to gain the morphological characteristics of BALB/c murine embryonic stem(ES) cells.The three techniques were compared and the results showed that each technique had its advantages.Therefore,the newer technique(AFM) should be combined with the classical ones(SEM and TEM).The normal hematopoietic stem cells(HSC) or CD34+ cells were observed by AFM.The AFM images of single HSC and the ultrastructure of HSC membrane were gained.Meanwhile we observed the ultrastructure of red blood cell membrane .Five parameters(Rp-v, Median height, Mean height, Rrms and Ra) were measured to quantitatively compare the characteristics of hematopoietic stem cell membrane and red blood cell membrane.The difference of aggregation behavior of type I collagen from different species was investigated by means of AFM.The results indicated that the aggregation behavior of collagen molecules was different under the same experimental conditions due to the difference of species.Our conclusions can be as reference for the development and improvement of collagen products.The imaging conditions of near-field optical microscope(NSOM) were explored Through comparison experiments.the effects of probes and feedback system on the quality of NSOM images were investigated.With different scan size,the transmission-mode and reflection-mode NSOM optical images were achieved of self-assembled collagen.The NSOM technique was also compared with AFM.
