泡桐体细胞胚胎发生及植株再生研究
Study on Plantlet Regeneration of Paulownia Plants Via Their Somatic Embryogenesis

【摘要】 泡桐是一种深受广大群众喜爱的速生用材树种,具有生长快、材质优良等特点,但其丛枝病和大冠低干等问题长期以来一直困扰着泡桐的进一步发展。随着现代科学技术的兴起,基因工程和细胞工程为解决泡桐的这些问题提供了有效的方法和途径。 离体植株再生研究是植物现代科学技术具体运用的重要基础。在植株再生的两种方式中,体细胞胚胎发生方式具有更大的优越性,它不仅可以作为细胞分化、形态发生等问题研究的理想体系,而且也是人工种子研制等的必要条件。 泡桐体细胞胚胎发生及植株再生体系的建立将为开展泡桐基因工程和细胞工程研究和培育泡桐新品种等工作奠定基础。本文通过对兰考泡桐、白花泡桐和毛泡桐无菌苗叶片和茎段体细胞胚胎发生及植株再生的研究,得出如下结果: 1.三种泡桐不同外植体细胞胚胎性愈伤组织的最适培养基分别为:兰考泡桐叶片为MS+0.3NAA+8BA,茎段为MS+0.3NAA+11BA;白花泡桐叶片为MS+0.3NAA+8BA,茎段为MS+0.3NAA+14BA;毛泡桐叶片和茎段都是MS+0.3NAA+17BA。 2.作为白花泡桐和毛泡桐杂交种的兰考泡桐在叶片和茎段胚性愈伤组织诱导条件敏感性等方面分别与毛泡桐叶片、白花泡桐茎段具有相似性;在体细胞胚胎发生规律等方面存在一定的差异。 3.不同种泡桐体细胞胚胎发生经历早期原胚、球形胚、心形胚和鱼雷胚等阶段最终达到成熟子叶胚。在实验条件下,泡桐体细胞胚胎发生以间接方式为主,其胚性愈伤诱导、各阶段体细胞胚胎发生以至体细 胞胚胎成熟和正常转换均在同一培养基同一培养条件下出现。
【Abstract】 Paulownia is a kind of popular, fast-growing trees which are of good quality,but these problems.such as witches broom, large crown and shout main stem obstruct Paulownia further denelopmento With the boom of modern science and technology, gene engineering and cell engineering provide effective methods to solve these problems.Research about the plantlet regeneration is basic for application of plants modern technology, between the two ways by which plantlets regenerate, somatic embryogenesis is more superior, it not only set up ideal systems for research about cell differeciation , morphogenesis etc, but also is necessary to study and make man-made seeds.Somatic embryogenesis and plantlet regeneration of Paulownia lay sound foundation for research on gene engineering, cell engineering and cultivation of new kind Paulownia, etc.having studied the somatic embryogenesis and plantlets regeneration from leaves and stem segments of Paulownia.tomentosa, Paulownia.elongata and Paulownia.fortunei, we come to such conclusions. 1. The optimum media for cell embyogenetic callus from differentexplants of three kinds of Paulownia respectively were: leaves of Paulownia.elongata MS+0.3NAA+8BA, stem segments of P.elongata MS+0.3NAA+11BA, leaves of P. fortunei MS+0.3NAA+8BA, stem segments of P.fortunei MS+0.3NAA+14BA, leaves of P.tomentosa MS+0.3NAA+17BA, stem segments of P.tomentosa MS+0.3NAA+17B A.2. Stem segments and leaves of P.elongata which is the progeny of P.fortunei and P.tomentosa were respectively similar to leaves of P.tomentosa and stem segments of P.fortunei in conditions under which embryogenetic callus were induced ,but to certain degree were different in tha law of somatic embryogenesis of different kinds of Paulownia.3. Somatic embryogenesis of different Paulownia experienced the same courses: early proembryo, globular embryo, heart-shaped embryo,torpe-do-shaped embryo and cotyledonary embryo. Under the experimental conditions ,the somatic embryogenesis of Paulownia mainly originated in indirect way. On the same media were found the induction of embryogenetic callus, embryogenesis of all phrases, even maturatiom and normal transformation of somatic embryo.
