三白草主要化学成分的分离检测及其对糖尿病的药效与药理研究

【摘要】 糖尿病是威胁人类健康的三大疾病之一。国内外学者都致力于研究有效治疗糖尿病的药物。中药是我国五千年文明的瑰宝。祖国医学积累了丰富的治疗糖尿病的史学资料,有许多治疗糖尿病的经典方剂。近年来,人们把中药治疗糖尿病的目标转向单味药物的筛选。三白草是新近发现的单味药物,有良好的降血糖作用。三白草起降血糖作用的化学成分的研究未见报道。通过此项研究,探讨三白草中起降血糖作用的化学成分以及它们的作用机理,为临床应用和进一步研究三白草治疗糖尿病提供客观的实验和理论依据,并为新药开发打下基础。 我们采取如下的方法进行此项的研究: 1.建立四氧嘧啶型糖尿病动物模型,探讨建立此模型的最佳实验条件。 2.对四氧嘧啶型糖尿病鼠一次和连续多次喂养三白草水煎剂,测出其体内超氧化物歧化酶(SOD)、丙二醛(MDA)、血糖水平的变化,以便判断三白草对四氧嘧啶型糖尿病动物的治疗效果。 3.三白草中提取出的几种化学成分喂养四氧嘧啶型糖尿病动物,检测它们对其血糖和超氧化物歧化酶(SOD)和丙二醛(MDA)的影响,用来判断起治疗效果的三白草化学成分。 实验结果显示:四氧嘧啶型糖尿病动物模型造模成功后,动物体内的超氧化物歧化酶(SOD)明显下降,此酶保护细胞免受损伤的能力降低;MDA值明显提高,脂质过氧化作用增强,间接反映细胞损伤程度增大。三白草水煎剂对四氧嘧啶糖尿病鼠一次给药和连续多次给药有明显降血糖作用,给药后3小时开始出现持续的降血糖作用,并持续8小时以上。单次和连续多次给药亦可明显提高动物体内的超氧化物歧化酶(SOD)的抗氧化能力,降低MDA脂质过氧化作用。经过从三白草中提取出的总黄酮、总氨基酸、多糖治疗后,四氧嘧啶型糖尿病兔体内SOD活性增强,MDA下降。 从以上实验实验结果可以得出如下结论:从三白草提取的化学成分中,总黄酮、总氨基酸和多糖,能有效降低血糖、降脂、提高超氧化物歧化酶(SOD)的活力、降低过氧化脂质及丙二醇、调节胰岛素分泌水平。说明三白草及其有效成分对预防和治疗四氧嘧啶型糖尿病起积极作用,在糖尿病并发症、胰岛素抵抗治疗上有一定的优势。

【Abstract】 Diabetes is one of the three most serious illnesses that greatly affect human’s health in modern society. Many efforts have been made on the research of medicines that can control and cure this illness. Kinds of classical medicines can be found from the history materials resulting from the abundant experience of diabetes therapy using Traditional Chinese Medicine(TCM). Recent year’s aim of treating diabetes by TCM has been turned to the selection of single-medicine to have a better effect. Saururs is a kind of single-medicine which was found recently having a good effect on reducing the blood-glucose, but no paper about its functional chemical components has been reported. This research focuses on Saururus’ functional chemical components of reducing the blood-glucose and studies the mechanism medicine behind its function to provide data for further study and clinical practice, and makes the foundation for the new drug exploitation. In this research includes:1. Establish Alloxan type diabetes model to investigate the best experimental conditions of such model. 2. Measure the continually change of Superoxide dismutase(SOD), methane dicarboxylic(MDA) and Blood Glucose level by multi-feeding of Saururas decoction to judge the effect of therapy on alloxan type diabetes rats. 3. Measure the change of SOD, MDA and Blood Glucose level by feeding alloxan type diabetes rats with different components extracted from Saururus to find the effectual chemical components.With successful modeling of alloxan type diabetes, we see a significant decrease in SOD level while MDA level increases, which hints a more serious damage to cell. Saururus decoction takes into effect on reducing the blood Glucose after 3 hours of feeding and such effect can last for more than 8 hours. It helps to lift the anti-oxidation ability of SOD and reduce the MDA lipid’s oxidation. After treating with the total flavone, total amino acid and polysaccharide extracted from Saururus, the increase of serum SOD and the decrase of MDA level in the alloxan-induced rabbits were observed.This research shows, Total flavone , total aminophenol and polysaccharide extracted from saururus can decrease blood-glucose(GLU), cholestero, increase SOD and decrease MDA, adjust the level of insulin. Saururus and its effectual components can be used for prevention and cure of tetrpyrimidinetype diabetes. Its effect on improving complication of diabetes is better than that of western medicine.
