木姜子组织培养技术体系研究

【摘要】 本研究以山苍子(Litsea cubeba Lout.Pers.)、四川木姜子(Litsea pungens Hemsl)、杨叶木姜子(Litsea populifolia Hemsl.Gamble.)的茎段为外植体进行组织培养技术体系的建立。着重研究木姜子离体培养的最佳培养基配方及培养程序,不同种的培养反应,以及工厂化育苗中有关简化培养及继代培养中交叉污染防止的措施。结果表明: 初代培养中以MS+BA2.0mg.l-1%IBA0.5mg.l-1为最佳组合培养基配方。依此配方确定11月中旬为取材的最佳季节,山苍子为首选树种(启动率90.3%、增殖数3.3、污染率30.3%),0.1%升汞对外植体处理时间4+4分钟效果较好。 继代培养,培养基为:MS+BA2.0mg.l-1+IBA0.1mg.l-1。其增殖系数高达7.7,丛生芽多且生长适中,30天可增殖一代。年理论增殖系数6.6×107。继代10次以后,用改良MS或B5代替MS可以使继代材料诱导分化恢复正常。防止交叉污染,用500mg.l-1浓度的硫酸链霉素和500mg.l-1浓度的硫酸庆大霉素抑菌效果较好。 生根培养,瓶内生根培养基为:1/2MS+IBA0.5mg.l-+1g.l-1活性炭。以纯砂和珍珠岩1:1混合的基质成活率最高(52.0%),瓶外带菌生根移栽用500mg.l-1ABT溶液对插条进行基部速蘸8秒钟,插入纯砂中,生根效果较佳。 简化培养程序,在污染率允许范围内(一般5%)可选用已煮沸过的自来水代替蒸馏水。用100℃沸水煮5分钟空培养瓶代替高压灭菌操作,可达到同样效果。

【Abstract】 The stems of three breeds of Litsea cubeba,Litsea pungens and Litsea populifolid were used for explant to establish the technique system of tissue culture. To find out the best medium ,proceeding,breeding reaction of species of Litsea in vitro culture. And to find out effective measures to prevent the pollution and simplification methods. The results were as follows:Initiation medium:MS+BA2.0mg.r’+IBA0.5 mg.l-1. Mid November was the best season to get material. The first best species Litsea cubeba ,the result 4+4 minutes was good to treat the explant by HgCl2(0.1%).Subculture medium:MS+BA2.0 mg.r’+IBA0.01 mg.l-1 .The rate of propagation could reach about 7.7 for 30 days. After ten times ,the induction and differentiation came back normally,replacing MS by B5 and modified MS.Rooting medium:1/2MS+IBA0.1 mg.l-1+ AClg.l-1 Out bottle rooting,the lower part of stems was soaked in ABT 500 mg.l-1 solution for 8 seconds ,then cut in medium containing only sand.The procedure of simplified educates:at the permission range of the rate of pollution (5%).the boiled tap water replaced distilled water,the boiled bottle replaced the high pressure sterilization.
