【摘要】 本研究从分子生物学的研究成果说明了恶性肿瘤产生的原因，以及抑制肿瘤的途径。其中，诱导肿瘤细胞凋亡是很重要的一环。并从西医药诱导细胞凋亡的原理引出了诱导细胞凋亡的理论依据和药理学原理。而现有文献证明，所有这些途径：致DNA损伤、抑制拓朴酶、抗代谢、激素样作用、诱导分化等，是不同类中药或方剂所具备的，这为中药诱导细胞凋亡提供了科学依据。 从祖国医学对肿瘤的认识、现代医家的肿瘤学说都支持扶正与祛邪是中医治疗肿瘤的主要原则这一认识。并且，由于中药药理学的创立，为在这一思路下，探讨在临床中发现的有抑瘤作用的药物的作用机理，提供了进行体外实验的较可靠的技术保证。 临床研究显示，六神丸、六君子汤在治疗肿瘤中分别以祛邪、扶正见长，收到良好效果。为进一步阐明它们的作用机理，故设计了“六神丸、六君子汤抑瘤、诱导肿瘤细胞凋亡的实验研究”。从动物体内实验、诱导肿瘤细胞凋亡的体外实验两方面，探讨它们的作用机理与细胞凋亡之间的关系。为提高临床用药的疗效提供依据。 实验结果显示，两者都具有抑制肿瘤的作用。但六神丸对荷瘤小鼠的作用以“祛邪”为主要功能方面，具有较高的诱导凋亡率，20％六神丸含药血清处理48h凋亡率为13.27％。方中麝香、冰片、蟾酥三味药物均属芳香之品，具有辟秽的功能，可能与其抑制肿瘤在生长过程中分泌出来的“毒素”物质损害人体方面发挥了作用有关。在运用六神丸+DDP治疗的同时，要辨证辅用扶正药物，以提高生存质量。六君子汤对荷瘤小鼠的作用以“扶正”为主要功能方面，诱导凋亡率较低，20％六君子汤含药血清处理48h凋亡率为7.97％。但在体重方面增重较多，且一般情况最好。提示：这可能与其在“内环境”条件下能调动扶正固本药诱导肿瘤细胞凋亡的免疫学机制，特别是提高其中的细胞免疫的免疫应答作用有关。更多还原

【Abstract】 From the angle of molecular biology the pathologic mechanism of malignant hyperplasia and the methods of suppressing tumor formation were introduced. The theoretical foundation and the fundamentals of pharmacology of the induction of cell withering that was the key to suppress tumor were expounded. Some Chinese herbs and formula made with herbs have effects of injuring DNA, suppressing topology amylase, anti-metabolism, hormone and inducing cell differentiation, which give scientific foundation for inducing cell to wither with Chinese herbs.According to TCM concept of wholism, tumor formation is due to the interaction between normal tumor precursor cells and the inner circumstance in human body that also effect on the existence and development of tumor, and canceration course is a potentially reversible course. Traditional Chinese medicine can indirectly adjust the existence and development of whole bodycells including tumor cell through adjusting the inner circumstance in the body.For the above-mentioned reasons, the experimental study of Liushen pill and Liujunzi decoction suppressing tumor and inducing tumor cell to wither was established, which including two parts of in vitro and in vivo to expound effectual mechanism of the two formulas and the relation between them and tumor cell withering.The results showed that both of the two formulas have effects on suppressing tumor. The main effect on mice carried with tumor of Liushen pill is to eliminate the pathogenic factor, which has a high rate of inducing tumor cell to wither. The withering rate for 48h of mice serum with 20% content of Liushen pill is 13.22%. Maybe, it is related to the effects of three drugs, Moschus, Verenum Bufonis and Borneolum, which suppress toxic substance secreted from tumor to injure body. Some drugs strengthening the body resistance should be used as Liushen pill associated with chemical therapy to treat tumor. The main effect on mice carried with tumor of Liujunzi decoction is to strengthen the body resistance, which has a less rate of inducing tumor cell to wither than that of Liushen pill. But Liujunzi decoction has a good effect on putting on weight of mice and ordinary physical condition of mice was better than those with Liushen pill. It implied that the two formulas coule improve body immunologic function to induce tumor cell to wither, especially to immune response in cellular immunity.更多还原
