树舌多糖GF对小鼠HepA瘤p53基因表达的影响

【摘要】 目的:为探讨GAPSGF对小鼠HepA瘤p53基因表达的影响,进一步完善GAPSGF抗瘤作用机制。方法:①抑瘤率:将各组完整的肿瘤组织称重,计算其抑瘤率。②应用ELISA法,观察GAPSGF对抑癌基因p53基因表达的影响;③通过SDS-PAGE,观察GAPSGF对HepA瘤血清蛋白组分的影响;④通过SDS-PAGE,观察GAPSGF对HepA瘤组织蛋白组分的影响;⑤通过DAGE法,来推测GAPSGF与PCD的关系。结果:①与阴性对照组比较,树舌多糖组和阳性对照组均有显著性差异(P<<0.05),其二者比较,无显著性差异(P>0.05)。②阴性对照组与空白对照组比较,二者有极其显著差异(P<0.01);树舌多糖组、阳性对照组与阴性对照组比较,差异均极其显著(P<0.01),接近于正常对照组(P>0.05);树舌多糖组与阳性对照组比较,无显著性差异(P>0.05)。③在Mr115kd处,较正常对照组,阴性对照组浅染,树舌多糖组和阳性对照组能使该带恢复正常。④a:在Mr65kd处,比较正常对照组,阴性对照组深染明显;树舌多糖组能使之变浅;阳性对照组对此无明显改变;b:在Mr53.7kd处,正常对照组呈浅染的条带,而阴性对照组呈深染的区带,GAPSGF呈浅染带,接近于正常对照组,阳性对照组也呈浅染带,但不能使之恢复正常;c:在Mr12.6kd处,正常对照组、阴性对照组、阳性对照组均无显著差异,但树舌多糖组在该处呈现深染区带。⑤正常对照组和阴性对照组均呈高浓度的单一条带,而树舌多糖组和阳性对照组电泳条带呈现大约200bp整数倍间隔的梯形条带。结论:①GAPSGF对HepA瘤有明显的抗瘤作用。②GAPSGF能明显减弱肿瘤组织中突变型p53基因的表达,且使P53含量接近于正常对照组。③GAPSGF可使HepA瘤血清异常蛋白组分恢复正常;④GAPSGF可使HepA瘤某些异常蛋白组分恢复正常;⑤GAPSGF有可能促进肿瘤细胞凋亡的作用。

【Abstract】 AIM: To study the influence of GAPSGF on p53 gene expression in HepA tissue. This will help to complete the mechanism of antitumour effect of GAPSGF. METHODS: ﹖he ratio of antitumour: Weigh the intact tumours of each group and calculate the ratio. ㎡bserve the effect of GAPSGF on p53 gene expression by the method of ELISA. ㎡bserve the effect of GAPSGF on the serum protein components by SDS-PAGE. (4) Observe the influence of GAPSGF on the tumour tissue protein components by means of SDS-PAGE. 〤onclude the influence of GAPSGF on PCD by DAGE. RESULTS: 〤ompared to Negative Control Group, both GAPSGF Group and Positive Control Group have notable difference (p<0. 05) , and the two , comparing to each other, have no difference (p>0. 05) .?There is much obvious difference between Negative Control Group and Norm Control Group (p<0. 01); Comparing GAPSGF Group and Positive Control Group with Negative Control Group, they have also much significant difference (p<0. 01) and they are similar to Norm Control Group (p>0. 05) , there is no difference between GAPSGF Group and Positive Control Group(p>0. 05). 〤ompared to Norm Control Group, Negative Control Group is understained at the band of Mr 115kd in the serum SDS-PAGE map, both GAPSGF Group and Positive Group can make it return. (4)In the tumour tissue SDS-PAGE map, a: In comparison to Norm Control Group, Negative Control Group is dark-dried ; GAPSGF Group can make it light, but Positive Control Group has no change to it at the band of 65kd. b: There is a more hyperchromatic band in Norm Control Group than Negative Control Group at the band of Mr 53. 7kd, GAPSGF can make it understained and similar to Norm Control Group. Positive Control Group can also lighten it, but can’ t resume it. c: At the band of Mr 12. 6kd, there is no notable difference among Norm Control GroupN Negative Control Group and Positive Control Group, but it is deep-dried in GAPSGF Group. ㊣n the DAGE map, a single$f GF jft/h m HepA ?p53hyperchromatic broad band is showed in Norm Control Group and Negative Control Group, but staircase bands , which are spaced by the euplidy of 200bp , are showed in GAPSGF Group and Positive Control Group. CONCLUSIONS: 〨APSGF has obvious antitumour effect on HepA tumour. 〨APSGF can obviously attenuate the expression of p53 mutation gene in HepA tumour and make P53 similar to Norm Control Group . (3) GAPSGF can resume the abnormal serum protein component. can resume some tumour abnormal tissue protein components. can probably accelerate HepA apoptosis.
