益心康对柯萨奇B3m病毒性心肌炎小鼠作用的实验研究

【摘要】 目的:本实验研究应用实验性病毒性心肌炎小鼠模型,观察中药复方制剂益心康对病毒性心肌炎小鼠一般状态的影响、对小鼠心肌细胞保护作用和对心肌细胞内病毒复制的抑制作用,初步探讨该药治疗急性病毒性心肌炎的作用机制,以寻找治疗病毒性心肌炎的新药,为临床提供一种有效的方法。方法:选用雄性Balb/c纯系小鼠75只,5-6周龄,体重19±2g,随机分为5组,包括中药治疗组、西药治疗组、盐水治疗组、盐水对照组、空白对照组,每组15只。中药治疗组、西药治疗组、盐水治疗组分别以含有柯萨奇病毒B3亲心肌株(CVB3m)的MEM Eagle氏液腹腔注射,诱导Balb/c小鼠形成病毒性心肌炎模型,并分别予益心康、病毒唑稀释液、生理盐水灌胃一周;盐水对照组予腹腔注射同等剂量不含病毒的Eagle氏液,生理盐水灌胃一周;空白对照组不予腹腔注射和灌胃。各组动物的饲养条件相同。动态观察各组小鼠一般状态的变化包括体重,造模一周后应用贝克曼全自动生化分析仪测每个小鼠血清磷酸肌酸激酶(CK)及其同功酶MB(CK-MB)。每组再随机抽取7只小鼠,并取这些小鼠的全部心肌,测心肌细胞感染CVB3mRNA总量。结果:造模3天以后,非造模各组小鼠一般状态明显好于造模各组,但中药治疗组小鼠一般状态好于其它两个治疗组。造模一周后造模各组小鼠平均体重显著下降,但中药治疗组小鼠的平均体重显著高于其它两个治疗组。造模一周后,造模各组小鼠的血清CK及其CK-MB和非造模各组相比显著升高,但中药治疗组小鼠的血清CK和CK-MB明显低于其它两个治疗组。造模一周后中药治疗组小鼠的心肌细胞含有的病毒RNA平均量明显少于其它两个治疗组,西药治疗组小鼠的心肌细胞含有的病毒RNA的量显著低于盐水治疗组。结论:益心康对柯萨奇B3m病毒性心肌炎小鼠有明显的生存保护作用,可以减轻心肌炎小鼠心肌细胞的损伤,并抑制柯萨奇病毒B3m在小鼠心肌细胞中的复制。
【Abstract】 Object: The experimental study selected the animal model of experimental viral myocarditis in Blab/c mice in order to study the effects of Chinese medicine Yi-xinkang in general state on these mice, in myocardial protection and in inhibition of viral replication in myocardial cells. Mechanism of function that the medicine treated viral myocarditis was preliminarily understood so that the new medicine could be looked for to treat viral myocarditis and a kind of effective way could be offered to clinical practice. Method: 75 male Balb/c purebred mice ,5 to 6-week-old , weigh49?g , were randomly divided into 5 groups : Yi-xinkang group (YI), Ribavirin group(RI), normal saline group(NS) , normal saline control group(NSC) and blank control group(BC) . Every group had 15 mice. The mice of YI, RI and NS were infected by coxsackie virus B3m (CVB3m) through abdominal cavity injection to develop animal models of experimental myocarditis in Balb/c mice, and were treated respectively by Yi-xinkang, Ribavirin and normal saline for a week. The mice of NSC were infected by MEM Eagle liquid without virus and treated by normal saline for a week. The blank control group wasn’t infected and treated. The other feeding conditions were same. Ordinary conditions of mice in every group were dynamically observed including their weight. CK and CK-MB of serum in each mouse were measured through Beckman automatic biochemistry analytical instrument after the mice were infected a week. 7 infected mice were randomly collected from every infected group, and amount of CVB3m RNA was measured from their myocardium. Result: After the mice were infected 3 days, general state of the infected groups was obviously worse than the non-infected groups, and general state of YI group was the best in three infected groups. Meanwhile, average weight of the infected groups was decreased, and average weight of YI was the highest in three infected groups. After the mice were infected 7 days, CK and CK-MB of serum in the infected groups were more increased than in non-infected groups, and those in YI were the lowest in three infected groups. Meanwhile, average of CVBsm RNA in the myocardial cells of YI was the lowest in the three groups. Conclusion: The results of the experiment suggest that Yi-xinkang might play an important role in protection of CVBsm viral myocarditis in the Balb/c mice, while it might reduce harm of viral myocardial cells in the infected mice and inhibit CVB3m replication in myocardial cells.
