葛根素提取分离工艺中试研究

【摘要】 葛根是一种既有丰富营养,又有独特药理作用的药食两用资源,用途广泛。作为葛根的主要活性成分,葛根素以其对心血管疾病的良好治疗作用,日益受到广泛关注,其高纯产品在国际市场上倍受青睐。 作者对提取分离工艺路线进行了合理设计,着重研究了葛根素提取分离中溶剂萃取和大孔吸附树脂吸附分离两步关键工序,筛选出了较优的工艺参数,并经中试放大试验验证,从而为葛根素的工业化生产工艺设计提供了重要参考依据。研究表明:萃取剂种类、原液pH值、无机盐种类与吸附树脂种类、树脂用量(干膏树脂比)、原液pH值、洗脱剂种类、流速等分别是两步工序中影响产品纯度和得率的主要因素;溶剂萃取的较优工艺参数为:以正丁醇为萃取剂、原液pH值为4-5、添加4%的醋酸铵;吸附分离的较优工艺参数为:选用AB-8型大孔吸附树脂、干膏树脂比为1:25、原液pH值为5、洗脱剂为30%乙醇、流速为1.5BV/h并进行梯度洗脱:利用此工艺参数生产葛根素具有产品纯、得率高、能耗低、操作简便等优点。
【Abstract】 Puerarin is a kind of resource for drug and food since it can provide people with a lot of nutriments and particular medicative effects. It has many uses. As the main active component of pueraria , puerarin is paid more and more attention increasingly for its good therapeutic effects to cardiovascular diseases, the pure product is very popular in international market.In this paper, the technology route of puerarin’s extraction and separation is designed rational1y.Two key steps such as solvent extraction and rnaroreticular resin absorption and separation are studied. At the same time, the optimal operation parameters are choosed and made sure through magnifying experiments. They can provide people with necessary references for the technology-designing of puerarin s industrial production.The experimental results indicate that the sort of the extracting solvent, pH of the original solution, the sort of salt and the sort of maroreticular resin, the dosage of it(the ratio of dry extract to resin),pH of the original solution, the sort of the eluent, the velocity of flow of it are respectively the main factors which influence the purity and obtaining ratio in two steps. The optimal working parameters in solvent extraction are listed as follows:the extracting solvent is 1-butanol;the pH of the original solution is 4~5;the salt is NHiOAc. Those in maroret icular resin absorption and separation are listed as follows:the maroret icular resin AB-8;the ratio of dry extract to resin is 1:25; the pHof the original solution is 5;the eluent is 30% ethanol; the velocity of flow is 2.5ml/min and applying gradient elution to this experiment.Using these operation parameters, a higher purity and obtaining ratio and a lower energy consumption, a easier operation can be obtained in puerarin’ production.
