GX4-1菌利用鱼粉废水产生絮凝剂的应用研究

【摘要】 本论文利用本实验室从活性污泥中筛选分离出来的絮凝剂产生菌GX4-1为菌种,考察了其以鱼粉废水、肉联厂废水、豆腐废水、熟肉制品厂废水、蟹足棒生产废水、新源玉米废水VTBR出水为替代培养基产生絮凝剂的情况,实验证明,该菌株单纯利用鱼粉废水就可以产生高效的生物絮凝剂。 菌株利用鱼粉废水生产絮凝剂的最佳培养条件为:将鱼粉废水稀释至COD浓度为10000~20000mg/L左右,调节pH为7~9,以10%的量接种,在温度为30℃、摇床转速为150~200rpm范围内培养3天。在此条件下培养得出的絮凝剂对高岭土悬浊液的絮凝率可高达99%以上。 起主要絮凝作用的物质存在于培养液中,菌细胞本身的絮凝作用不大。 即使不对培养基灭菌,该菌也能产生絮凝剂而且絮凝效果并不减弱。但灭菌时的絮凝剂粗品产量要略高于不灭菌时的絮凝剂粗品产量。这可能是由于该菌经活化后,活性很高,在接入发酵培养基后能够大量生长并迅速占据优势,因而并不对其生产絮凝剂构成影响。 实验证明,菌株GX4-1在利用鱼粉废水进行半连续培养生产絮凝剂是比较稳定的。同时还发现在对培养基灭菌和不灭菌两种情况下,该菌的生长情况和絮凝剂产生情况相类似。 GX4-1菌利用鱼粉废水产生的生物絮凝剂对高岭土悬浊液、土壤悬浊液、细活性炭悬浊液、碳素墨水以及电瓷厂废水等无机类废水有很好的除浊效果。对生活污水、重油催化废水、针织染纱废水和大化焦化废水等有很好的除浊功能和较好的COD去除效果。同时发现在偏碱性条件下该絮凝剂的絮凝效果较好。 GX4-1菌利用鱼粉废水产生的生物絮凝剂对鱼粉废水进行絮凝时,在絮凝条件为:10%的CaCl2溶液加量为15ml,微生物絮凝剂的添加量为30ml,pH为8的时候,其COD(原水的COD为80000mg/L左右)可以降至21600mg/L左右,COD去除率达到73%左右;其OD550可以降到0.22左右(原水的OD550远大于2)。此时发现沉降物中含有约48.5%的粗蛋白,因而可用来做饲料添加剂。
【Abstract】 Several kinds of wastewater as the culturing medium of the strain GX4-1,which was screened by us from activated sludge as the bioflocculant producing bacterium, was tested .We found that it can produce bioflocculant with high flocculating rate if we only used fish meal wastewater as the culturing medium.Under the optimum culture conditions: the COD(the concentration) of diluted fish meal wastewater about 10000~20000mg/L,pH 7~9,30癈, 150~200rpm,with the inoculation amount of 10 percent,cultivated for 3 days in flask,the flocculating rate of the bioflocculant to kaolin clay suspension can reach 99 percent.The main flocculating material was found in the culture broth,but not in the microbial cells.If the medium was not sterilized, the flocculating activity of the bioflocculant the strain produced was no less than that with medium sterilized.However,the yield on the former condition was a little less than that on the latter condition.Maybe the reason is that the bacterium could grow rapidly and occupy a dominant position in the system soon because it was inoculated with high activity after it had been activated.In experiments,we also found that the process of semi-continuous culture of the bacterium using fish meal wastewater as medium was steady.And on both the conditions of medium sterilized and not sterilized, the flocculating activity of the bioflocculant the strain produced was similar.The bioflocculant produced by strain 6X4-1 using fish meal wastewater show high flocculating rate to kaolin clay suspension, soil suspension,fine activate carbon powder suspension,carbon ink water and wastewater from electroceramics plant.And it also has a high ability to remove the turbidity of domestic sewage,catalyzed dead oil wastewater,dyeing wastewater in knitting plant and wastewater generated in the coking process in chemical plant and to degrade their values of COD. At same time,we found that the flocculating activity of the bioflocculant is higher when the coagulating system is in a little basic condition.As the adding volume of CaCl2 solution which mass concentration is 10% is 15ml and the doses of the bioflocculant is 30ml with pH about 8,the fish meal wastewater’s COD value decreased to 21,600mg/L with the initial COD value about 80,000mg/L for floculating by the bioflocculant.The degrading rate of COD reaches about 73%, and the OD550 of the wastewater could be decreased to 0.22 with the initial value of far beyond 2. At same time, about 48.5% crude protain was found in the sediment,and it may be used as feed additive.
