微生物转化法生产二羟基丙酮

【摘要】 本论文从来自于不同地区的50多份土壤中筛选到能转化甘油为二羟基丙酮的微生物茵株30多株,对各菌株二羟基丙酮的生产能力进行了鉴定、比较,并以其中转化活性最高的菌株delqh6为材料,考察了微生物发酵法与酶转化法生产二羟基丙酮的工艺条件及主要影响因素,论文主要由如下内容组成: 综述了二羟基丙酮的性质和用途,阐述了醋酸细菌属细胞中甘油的运输机制与代谢机理,概括了二羟基丙酮的微生物转化方法与有关微生物发酵法与酶转化法生产二羟基丙酮的工艺条件及主要影响因素;创立了甘油存在下二羟基两酮的定性、定量分析方法,完善了甘油的高碘酸定量分析方法,自行设计了菌种筛选思路与模型,设计了菌种筛选实验步骤,对自然界所采土壤样品进行菌种筛选,对所筛菌株二羟基丙酮的生产能力进行鉴定和比较,其中甘油到二羟基丙酮的最高转化率为16%,并以这一菌株(命名为delqh6)为试验材料既研究了微生物发酵法与酶转化法生产二羟基丙酮的工艺条件和主要影响因素,也研究了甘油脱氢酶的形成条件及其反应活性的主要影响因素,优化培养基配方及主要工艺条件,探索微生物发酵法与酶转化法生产二羟基丙酮的合适工艺条件,本论文实验中,甘油到二羟基丙酮的最高转化率达25%以上。 最后比较了两种二羟基丙酮的生产方法,并展望了酶转化法生产二羟基丙酮的潜在应用价值。
【Abstract】 More than 30 strains capable of producing Dihydroxyacetone from glycerol were isolated from fifty soil samples collected from different regions, then a comparison with all these strains concerning their converting capability of Dihydroxyacetone was made. Moreover, the paper has done much research on the literature of Dihydroxyacetone production based on delqh6, one of these above strains and the whole process of this paper is as follows: First, it summarized the characteristics and uses of Dihydroxyacetone, described glycerol transport mode together with its metabolic mechanism inside some acetic bacterium and reviewed the literature concerning both microbial fermentation and enzymatic production of Dihydroxyacetone. Then, the paper set up the quantitative and qualitative analytical method of Dihydroxyacetone in the presence of glycerol, revised the periodatic analytical process of glycerol, made a screening of Dihydroxyacetone? producing strains and moreover, it also compared all these strain isolated from soil samples by their ability of converting glycerol to 1,3? Dihydroxyacetone, among this experiments, the highest conversion rate from glycerol to Dihydroxyacetone is 16%. Then,using the strain delqh6 of the highest converting capability, the paper studied the literature on microbial fermentation production of Dihydroxyacetone and meanwhile, much analysis, on the formation of glycerol dehydrogenase together withits characteristics and on the literature for the enzymatic productionof Dihydroxyacetone, was done based on the strain de1qh6. The paperinvestigated the main influential parameters on the production ofDihydroxyacetone, by adjusting environmental conditions and by modifyingmush ingredients and it found a optica1 li terature condi tion forDihydroxyacetone production. Inside this paper, the highest conversionrate from glycerol to l, 3--Dihydroxyacetone is over 25%. At last, acomparison with the above two method of Dihydroxyacetone productionmentioned in this paper was made and furthermore, it estimated thepractica1 uses of enzymatic production in the industry.
