丽格海棠离体繁殖技术研究

【摘要】 本研究以丽格海棠的叶、茎、花茎、叶柄等营养器官作外植体进行组织培养,着重研究丽格海棠的最佳外植体、最佳培养基配方及培养程序,以及生根炼苗等的关键技术。研究表明初代培养的基本培养基为MS,激素配比为6-BA0.5mg/L+NAA0.1mg/L,培养30天可获得较多的不定芽。外植体分化能力的研究表明:从不同类型外植体分化能力来看,叶>茎>花茎>叶柄;从叶片的不同部位来看,叶中>叶侧>叶边缘;从不同叶龄来看,中等成熟叶>幼叶>完全成熟叶>芽叶。叶片的接种模式:反放比正放好。而传统的接种模式认为双子叶植物叶片的气孔主要分布在叶背,因而正放有利于养分和水分的吸收。本试验正放叶片的成活率很低,实验结果与传统的接种模式相反,因此气孔作为养分和水分的主要通道之说有待于进一步研究。继代培养培养基为:MS+6-BA0.05mg/L,30-40天可增殖一代,平均每代增殖系数大于5。瓶内生根培养:培养基为3/4MS+IBA1.0mg/L,生根率可达82.4%;瓶外生根以无根苗基部速蘸700mg/L的AST溶液8秒,生根效果较好,生根率达52.0%。炼苗基质以沙:蛭石:珍珠岩=1:1:1成活率最高,成活率可达61.25%。丽格海棠完整的组培技术体系在国内尚无成功的先例,本研究在丽格海棠初代培养,外植体的选择,继代培养,瓶内外生根炼苗等关键环节取得较大进展。本试验结果突破了叶片接种模式正放为好的传统理论,有较高的理论意义,在实践上有一定的参考价值。

【Abstract】 Study on the Technique system of propagatio in vitro in Rieger Begonia Li Chum Xiii (College of Forestryand Horticulture, Sichuan Agricultural University, Ya抋n,Sichuan,China,625014) ABSTRACT Leaf. stems~ stenisof flower~ leafstalk of Rieger Begonia were used for explants in study of organ culture to establish a rapid technique system of propagation in vitro,and to find out the best explant.. medium culture procedure and effective measures to improve the survival rate of seeding,The results were as follow: Iniation mediuntMS+6-BA0.5mg/L+NAAO. lmg/L can get more adventitious buds,afler 30 days culture. The study of differentiation capacity: From different kind of explant,leal5stem>stem of flowes>leafstalk From different section of leaf,niiddle of leaf >lateral of Ieaf>edge of leaf From different age of the leaI~ midium mature leaf >immature leaf>mature leaf >bud leaf Inoculation model: lay it up in the obverse side of the leaf ,the lower survival rate we get, so lay it up the reverse side better than the obverse sideof the leaf Subcultur medium:MS+6-BAO.O5mg/L, theoretically, the rate of propagation of Rieger Begiona could reach about 7 for 30-40 days. In bottle rooting medium: 3/4MS+IBAO. lmg/L, the rooting rate was about 800/o Out bottle rooting:the bottom of stems was soaked in ABT 700 mg/L solution for 7 seconds, then put into medium (sand: vermiculite: pearlitem=1:1:1).The effection is good, the rooting rate was about 52%. Acclimatization medium(sand:vermiculite:pearlite=l :1:1) the higher survival rate, it is about 61.25%.
