两个水稻品种抗纹枯病数量性状基因定位
Quantitative Trait Loci Controlling Sheath Blight Resistance in Two Rice Varieties

【摘要】 本实验以Lemont/特青的F2无体系群体为材料,其中99年每个系只设一个区组,2000年设两个区组,每组8个单株。以牙签嵌入法接种F2群体,以0-9级病级划分标准调查F2群体各个单株无性系的病级,对F2的143个无性系家系进行SSR(Simple Sequence Repeats)分析,以分群分析法(Bulked Segregant Analysis)构建抗感基因池,应用分子标记在抗感池中筛选阳性标记,然后将阳性标记上群体进一步检测和验证主效QTLs的存在。结果如下: 1、抗亲特青和感亲Lemont间存在着丰富的多态性。 共选用28个RFLP克隆探针和320个微卫星标记,其中21个RFLP探针和220个微卫星标记可揭示双亲间多态性,多态性揭示率为69%,说明本研究所用的杂交组合为遗传图谱构建和基因定位的理想材料。 2、杭稻纹枯病主效QTL区间作图分析 以水稻抗纹枯病为目标性状选择抗病和感病的极端个体来构建抗感等基因池,将已检测出的在两亲本间具有多态的探针或引物分别进入下一轮的抗感池的筛选,在两池间共检测到5个阳性标记(RM201,RM160,RM245,RM167,RM202),位于第9染色体或第11染色体。通过对阳性标记所在染色体区域构建微卫星遗传图谱,上群体验证,99年和2000年均在这两条染色体各发现一个QTL位点,分别定名为qSB-9、qSB-11,其中qSB-9两年均作图于同一个区间RM205-RM201之间;而qSB-11两年结果在相邻区间,分别在RM167-RM20B和RM202-RM167之间,但接近RM167。qSB-9两年分别解释表型变异的12.9%和15.3%,qSB-11两年分别解释表型变异的20.7%和11.6%。qSB-9来自特青,而qSB-11来自Lemont。 3、主要农艺性状的QTL作图分析 99年对株高和生育期进行了调查,2000年则又增加了有效穗数的调查,通过分子标记在已构建的连锁图谱上进行有关基因的定位。结果在第12染色体的RM247-RM101标记区间检测到一个株高QTL(99年人定名为 qH上,在第 7染色体上的 RMZI 4RMZ标记间检测到一个生育期 QTL(99.2000* 定名为 qHDJ 2000年在第 2染色体上还检测到一个有效穗QTL,定名为qEH上,这些QTLS对目标性状的贡献率分别为10.5%、11.5%和 7%。
【Abstract】 This study was conducted with a clonal F2 population from the cross between Lemont and Teqing .In 1999 each F2 population was planted into one block with 8 plants for each line while each were planted into two blocks in 2000.The population which includes 143 clonal families were inoculated with RH-9 fungus.The extreme lines were selected to construct resistance and susceptibility DNA pools, respectively, for bulked segregant analysis.The main results are summarized as below. 1. There were abundant polymorphisms between two parents. 28 RFLP probes and 320 microsatellite primes were used to detect polymorphism, 21 RFLP probes and 220 microsatellite primes showed polymorphism, the ratio of polymorphism between two parents was 69%. The result indicated that this cross was an ideal research material for genetic map construction and gene mapping. 2.lnterval QTLs analysis of resistance to rice sheath blight: In the present study, two major QTLs for rice sheath blight were identified in both years. The probes which revealed polymorphism between two parents were chosen for further analysis between two DNA pools, and five primers were discovered, three of them on chr 9 and the other two on chr 11 .The two QTLs were named qSB-9 and qSB-1 I respectively. qSB-9 was located in the interval between RM2OS-RM2Olin both two years .And qSB- 11 was located between RM 167 and RM2OB in 1999 and RM202 and RM167 in 2000,but both near to RM167,the middle primer .qSB-9 could explain 12.9% and 15.3% of phenotypic variation respectively and qSB-llcould explain 20.7% and 11.6% of phenotypic variation respectively 3.Interval QTLs analysis of some morphological characters: With the constructed map some other QTLs were located with SSR .4 primers. One QTL (qPH-12) controlling plant height was discovered in 1999, one QTL (qEH-2) controlling effective panicle in 2000, and the other QTL (qHD-7) controlling heading date was discovered in both two years. These QTLs could explain 10.5%, 7% and 11.5% of phenotypic variation, respectively.
