赣南脐橙皮橙皮苷与果胶的联产工艺研究

【摘要】 为了充分开发柑橘加工废弃资源的价值,本文以赣南脐橙皮为原料,探究橙皮苷和果胶联产工艺及其理化性质。通过单因素和正交实验得到橙皮苷和果胶联产最优工艺条件,通过高效液相色谱、傅里叶红外光谱、差示扫描量热仪、紫外光谱等仪器评价橙皮苷纯度及表征其结构,借助果胶酯化度、溶解度、pH值、红外光谱等来表征橙皮果胶的理化性质。结果显示,通过碱溶酸析法纯化橙皮苷,在料液比(g/mL)1:50、碱溶pH值12.5、提取时间30 min、酸析pH值5.0时,橙皮苷纯度最佳为96.79%,且得率为50.64%。在料液比(g/mL)1:20,pH值2.2,提取时间60 min,提取温度90℃的条件下,橙皮果胶得率最高,达22.38%,果胶中半乳糖醛酸含量为79.20%,酯化度为77.16%,胶凝度为153.3度,果胶酸不溶灰分为0.83%,pH值为2.79,以上指标表明该橙皮果胶为优质高酯果胶。

【Abstract】 To fully develop the value of waste resources in citrus processing, the co-production process of hesperidin and pectin,and the physicochemical properties of pectin were studied by using Gannan navel orange peel as raw material. Single factor and orthogonal experiments were used to determine the optimal process conditions for the co-production of hesperidin and pectin; the purity of hesperidin and its structure were evaluated by high performance liquid chromatography(HPLC), Fourier-infrared(FTIR)spectroscopy, differential scanning calorimetry(DSC), and ultraviolet(UV) spectroscopy. The physicochemical properties were studied by the esterification degree of pectin, solubility, pH value, FTIR spectroscopy and other indicators. The results indicated that the best purity of hesperidin was 96.79% and the yield was 50.64% when the solid-liquid ratio was 1:50, the alkali soluble pH value was 12.5, the extraction time was 30 min, and the acid precipitation pH value was 5.0. Under the conditions of the materialliquid ratio 1:20, pH 2.2, extraction time 60 min, and extraction temperature 90 ℃, the extraction rate of orange peel pectin was the highest, reaching 22.38%. The pectin contained 79.20% galacturonic acid, had an esterification degree of 77.16%, a gel strength of 153.3 degrees, an insoluble ash content in pectic acid of 0.83%, and a pH value of 2.79. These indicators suggested that this orange peel pectin is a high-quality high-ester pectin.
